摘  要:目的观察乳康饮对移植瘤裸鼠乳腺癌组织Ang-2、Ang-2mRNA表达的影响,探讨复方中药抑制乳腺癌转移的机制。方法 BALB/c裸鼠30只,乳腺原位种植人乳腺癌细胞株MDA-MB-435S,建立乳腺癌移植瘤自发性转移模型后随机分成6组,分别为疾病模型组,5-FU对照组,乳康饮低、中、高剂量组,乳康饮加5-FU组,每组5只裸鼠。疾病模型组给予生理盐水灌胃;5-FU对照组给予腹腔注射;乳康饮低、中、高剂量组分别给予乳康饮不同剂量灌胃;乳康饮加5-FU组给予5-FU腹腔注射,同时给予乳康饮中剂量灌胃,均每天1次,连续6周。用药后检测肿瘤直径和腋窝淋巴结转移抑制率,免疫组化、PCR技术分别检测乳腺癌组织Ang-2的表达,分析各组的表达差异。结果裸鼠乳腺原位均有实质性肿瘤生长及腋窝淋巴结转移,各药物组肿瘤直径与疾病模型组比较明显减小(P<0.05),各药物组比较差异无统计学意义(P>0.05);乳康饮+5-FU组抑瘤率明显高于其他各药物组(P<0.05)。各药物组对淋巴结转移均有抑制作用,乳康饮各剂量组的淋巴转移抑制率与5-FU组差异无统计学意义(P>0.05),但与乳康饮加5-FU组有显著性差异(P<0.01)。免疫组化结果显示Ang-2在疾病模型组的表达与乳康饮中、低剂量组差异无统计学意义(P>0.05),与乳康饮高剂量组、5-FU对照组、乳康饮加5-FU组比差异有统计学意义(P<0.01或P<0.05)。PCR结果显示疾病模型组裸鼠乳腺癌组织Ang-2mRNA的相对表达量均高于各药物组,差异有统计学意义,与乳康饮高、中、低剂量组比(P<0.05),与5-FU对照组、乳康饮加5-FU组(P<0.01)。结论乳康饮可能通过干预Ang-2蛋白和基因表达,干扰Ang-2/Tie2信号通路,抑制肿瘤组织的淋巴管生成,从而达到抑制乳腺癌淋巴转移的作用。Objective To observe the effect of Ruykanyin (RKY) on the expression of Ang-2,Ang-2mRNA in transplanted tumor of nude mice breast cancer tissue,discuss the inhibition mechanism of the traditional Chinese medicine compound Rukangyin in breast cancer metastasis.Methods Human breast cancer cell line MDA-MB-435S were in situ implanted into 30 BALB/c nude mice to establish breast cancer transplantation tumor spontaneous metastasis model.Then they were randomly divided into six groups,i.e.the disease model group,the 5-FU control group,the low,medium,high dose RKY groups and the RKY + 5-FU group,with 5 nude mice in each group.Physiological saline gastrogavage was given to the model group,peritoneal injection to the 5-FU control group,different dose gastrogavage to the low,medium,high dose RKY groups,5-FU peritoneal injection to the RKY + 5FU groups.At the same time,medium dose Rukangyin gastrogavage was given to the rats once a day for 6 successive weeks.The tumor diameter and the inhibition ratio of cancer tissue was detected by immunohistochemical technique and RCT technique,respectively.Axillary lymph node metastasis were detected after medication.The expression of Ang-2 in breast and the expression differences in each groups were analyzed.Results The growth of nude mice breast cancer and the axillary lymph node metastasis can be found in each groups.The diameter of the tumor was reduced significantly in each medicine groups compared with the disease model groups (P 〈 0.05).There was no statistically significant difference in each medicine groups(P 〉 0.05).The inhibition rate was significantly larger in the RKY + 5-FU groups compared with each medicine groups(P 〈 0.05).Each medicine group could inhibit lymph metastasis,and the inhibition rate of lymph metastasis in different dose RKY groups had no significantly difference compared with 5-FU groups(P 〉 0.05),but the inhibition rate of lymph metastasis in different dose RKY groups had significantly difference compared with RKY + 5-FU
