摘  要:目的探讨金属硫蛋白对镍铬联合染毒小鼠睾丸损伤的保护作用及其机制。方法利用硫酸镍和重铬酸钾联合染毒5 d复制小鼠睾丸损伤模型,然后腹腔注射不同剂量的金属硫蛋白7 d,在36 d处死小鼠,检测睾丸脏器系数、睾丸结构,精子数量、活力和畸型;检测睾丸组织中MDA含量、SOD、LDH和SDH活性。结果MT保护组小鼠睾丸脏器系数、精子数量及精子活动率随MT剂量增大而增加,精子畸形率随MT剂量增大而降低,各MT保护组与模型对照组比较差异均有显著性(P<0.05),并有较好剂量依赖关系;模型对照组小鼠睾丸组织结构损伤严重,MT组小鼠睾丸损伤较对照组轻,MT剂量越大,睾丸损伤越轻;随MT剂量增加,MT保护组小鼠睾丸SOD、LDH和SDH的含量逐渐增加(P<0.05),MDA含量逐渐降低(P<0.05),均有较好的剂量依赖关系。结论金属硫蛋白对镍铬联合染毒小鼠睾丸损伤有保护作用,保护机制可能与MT促进镍铬排出、减轻氧化损伤及增加LDH、SDH的活性有关。[ Objective ] To explore the protective effect and mechanism of metallothionein to mice testis damaged by nickel and chromium. [Methods] The mice testis injury model induced by nickel and chromium was injected different dose metallothionein into abdomen for consecutive 7 days, once a day and 0.1 mL/10 g body weight. All mice were sacrificed in the 36th day. The testicular organ coefficient, sperm quantity, vivi-sperm rate, malformation rate and the testicular tissue morphous changes were observed. The concentration of SOD and MDA and the activity of LDH and SDH were detected in the testes. [ Results ] In the metallothionein groups, the testicular organ coeffi- cient, sperm quantity, vivi-sperm rate, and the activity of SOD, LDH and SDH were gradually increased with the in- creasing of metallothionein dose (P 〈0.01), and malformation rate and the content of MDA were gradually decreased (P 〈0.01) compared with testis injury model groups. And also there was dose-effect relationship (P 〈0.05). The de- gree of mice testis injury was lighter in the metallothionein protected groups than testis injury model group (P 〈0.05). [ Conclusion] The metallothionein can protect mice from testis injury induced by nickel and chromium, and the protective mechanism may be related to reduce oxidative damage, increase the activity of LDH and SDH and pro- mote excretion of nickel and chromium in the testis.
