摘 要:目的:研究松茸醇提部位(MTX)对Hela细胞的生长抑制作用及其作用机制。方法:采用四甲基偶氮唑蓝(MTT)比色法检测MTX对Hela细胞的增殖抑制率;用流式细胞仪分析细胞周期的变化;通过免疫荧光技术检测MTX对微管聚合的影响;并用蛋白免疫印迹法检测微管蛋白表达的变化。结果:MTX能明显抑制Hela细胞的增殖,抑制率呈剂量和时间依赖性,能使Hela细胞的G2/M期比例逐渐升高,免疫荧光结果表明,MTX作用后能引起Hela细胞微管网络异常,蛋白免疫印迹结果表明α-tubulin和MAP2的表达显著下降。结论:MTX具有抑制Hela细胞的增殖作用,其机制可能与其降低微管蛋白的表达,抑制细胞微管聚合有关。OBJECI'IVE To investigate the anti-tumor effects of MTX on the growth of Hela cells and to elucidate the possi ble mechanism. METHODS Hela cells were treated with certain concentrations of MTX. and the inhibitory rates on growth were determined by MTT assay. Flow cytometry was performed to measure the cell cycle arrest and immunofluorescence was used to detect the tubulin polymerization. Western blotting was performed to determine the expression of microtubule protein. RESULTS MTX inhibited the growth of Hela cells in a dose and time dependent manner. The percentage of Hela cells in G2/ M phase was increased and the microtubule polymerization was inhibited. The inhibitory effect of MTX on expression of a-tu- bulin and MAP2 protein increased as a time-dependent manner. CONCLUSION MTX inhibited the growth of Hela cells associ ated with down regulated the expression of microtubule protein and inhibited the microtubular polymerization.
