摘 要:试验旨在研究苜蓿皂苷对山羊瘤胃原虫体外发酵功能的影响。选择4只体况良好、安装永久性瘤胃瘘管的徐淮白山羊,晨饲后3 h采集瘤胃液,分离出原虫,采用体外批次培养设计进行体外发酵。根据苜蓿皂苷的添加量分为4个处理组:对照组不添加(0),3个试验组分别添加1%、2%及3%的苜蓿皂苷,每组3个重复,体外厌氧培养24 h后,分别测定pH值、氨态氮(NH3-N)浓度、微生物蛋白(MCP)浓度及挥发性脂肪酸(VFA)含量。结果表明:添加1%~3%的苜蓿皂苷对瘤胃原虫培养液的pH值、NH3-N浓度均无显著影响(P>0.05),但降低了乙酸、丙酸、丁酸和TVFA浓度,且乙酸/丙酸有升高趋势(P>0.05);此外,2%和3%的苜蓿皂苷极显著降低了培养液中MCP浓度,较对照组分别降低24.5%和17.1%。提示苜蓿皂苷对瘤胃原虫有一定的抑制作用,且对原虫发酵产生丙酸的抑制效应大于乙酸。This research was conducted to investigate the effect of alfalfa saponins on ruminal protozoa1 fermentation in vitro culture. Four ruminally-fistulated XuHuai goats were used in the experiment. Ruminal fluid was collected at 3 hours after morning feeding and fermented in vitro when the protozoal was isolated from it. The addition levels of alfalfa saponins were 0 (control group), 1%, 2% and 3% respectively. Each group has 3 replicates and anaerobic acculturated for 24 h. The results showed that adding 1%-3% alfalfa saponins did not affect the pH value or the concentration of NH3-N (P〉0.05), but decreased the concentration of acetate, propionate, butyrate and TVFA, and the acetate/propionate ratio had an increasing trend (P〉0.05). What's more, the concentration of MCP was extremely decreased by 24.5% and 17.1% in 2% and 3% group when compared with the control group. Results indicated that alfalfa saponins could inhibit the ruminal protozoal fermentation, and the inhibitive effect was more intensive on propionate than acetate.
