摘　　要：目的:探讨纤维蛋白原样2(fibrinogen-like protein 2,fgl2)在重症急性胰腺炎(severe acute pancreatitis,SAP)大鼠胰腺中的表达及意义。方法:48只SD大鼠随机均分为假手术(sham operation,SO)组及SAP组。实时荧光定量PCR法、免疫组化法检测fgl2在大鼠胰腺及外周血单个核细胞(peripheral blood mononuclear cell,PBMC)中的的表达。结果:与SO组相比,fgl2在SAP大鼠胰腺微血管内皮(P<0.01)及PBMC(P<0.05)中高表达;SAP组胰腺病理评分显著高于SO组(P<0.01),且与胰腺(r=0.852,P<0.01)及PBMC(r=0.735,P<0.01)中fgl2表达呈正相关。结论:fgl2在胰腺及PBMC中的表达与胰腺损伤程度密切相关,因此可能可用作早期预测SAP发生的标记物。Objective： To probe the expression of fibrinogen-like protein 2 （fg12） in rats with severe acutepancreatitis （SAP） and to explore its significance in the development of SAP. Methods： Forty-eight Sprague- Dawley rats were randomly divided into two groups： sham-operation （SO） group and SAP group. 4% sodium taurocholate were applied by retrograde injection into rats＇ biliopancreatic ducts to induce SAP. Specimens of 1 h, 4 h, and 8 h since induction were collected; HE staining was carried out to observe the pathological changes in pancreatic tissues; the expression of fg12 prothrombinase in both pancreatic tissues and peripheral blood mono- nuclear cells （PBMCs） were examined with real-time PCR and immunohistochemistry. Results： Pathological changes of the pancreas confirmed the establishment of SAP model. Levels of fg12 mRNA were evidently elevated and proved to be significantly different in pancreas （P〈0.01） and PBMCs （P〈0.05） in rats that developed SAP compared to those in SO group; the mean pathological score in SAP group was significantly higher （P〈0.01） than that in SO group, and closely correlated with fg12 expression both in pancreatic tissues （r=-0.852, P〈0.01） and PBMCs （r=0.735, P〈0.01）. Conclusion： The up-regulated fg12 expression is correlated with the severity of pancreatic injury closely, thus may predict the occurrence of SAP as a useful biomarker on the onset of the disease.
