摘　　要：目的探讨MCPH1(microcephalin 1)基因过表达对人肺癌A549细胞迁移及侵袭能力的影响。方法将质粒pcDNA3.1(-)/MCPH1(实验组)和pcDNA3.1(-)(阴性对照组)分别转染人肺癌A549细胞,采用Real-time PCR及Western blot法分别检测转染48 h后的细胞内MCPH1的表达;采用细胞划痕试验及Transwell试验分别检测MCPH1过表达对细胞迁移及侵袭能力的影响。结果实验组A549细胞中MCPH1水平明显高于阴性对照组,差异有统计学意义(P均<0.05);实验组细胞迁移及侵袭能力较阴性对照组明显降低,差异有统计学意义(P均<0.05)。结论 MCPH1基因过表达可明显抑制A549细胞的迁移及侵袭能力。Objective To investigate the effect of microcephalin 1 （MCPH1） gene overexpression on invasion and migra- tion abilities of human lung cancer A549 cells. Methods A549 cells were transfected with plasmid peDNA3. 1 （-） / MCPHI （test group） and peDNA3. 1 （-） （negative control group） respectively, and determined for expression of MCPH1 by Real-time PCR and Western blot 48 h later. The effects of MCPH1 overexpression on invasion and migration abilities of A549 cells were evaluated by wound healing test and Transwell test respectively. Results The MCPH1 level in A549 cells in test group was significantly higher than that in negative control group （P 〈 0. 05 ）. However, both the migration and invasion abilities of A549 cells in test groups decreased significantly as compared with those in control group （P 〈 0. 05 ）. Conclusion The overexpression of MCPH1 gene significantly inhibited the invasion and migration of A549 cells.
