摘  要:研究不同室温对小鼠过敏性哮喘模型脾脏、外周血细胞共刺激分子的表达及肺组织损伤的影响,探究温度与过敏性疾病严重程度的相关性。将40只昆明鼠随机分组后分别置于20±2℃(低温组)和30±2℃(高温组)饲养,每个温度组分别设置造模组和对照组。造模组小鼠分别于第1天及第8天腹腔注射致敏液0.5ml(含卵清蛋白1 mg和氢氧化铝凝胶4mg),第15天起,每天以1%卵清蛋白溶液喷雾30min,连续7d,末次喷雾18h后处死小鼠。对照组用相同体积的生理盐水处理。免疫荧光及流式细胞术检测脾脏和外周血PBMC共刺激分子的表达,ELISA检测血液中IgE、IL-4及IFN-γ的含量,取肺组织经HE染色后作组织形态学分析。造模组脾脏和外周血PBMC共刺激分子的表达均升高(P<0.05),且低温组明显高于高温组(P<0.05)。造模组血清IgE及IL-4的水平明显升高(P<0.05),低温组明显高于高温组(P<0.05);但IFN-γ的水平明显下降(P<0.05),两温度组无明显差别(P>0.05)。组织形态学的观察结果显示,造模组小鼠的支气管及肺泡腔内出现炎性细胞浸润、支气管管壁增生及肺泡黏膜肿胀水肿,且低温组比高温组严重。哮喘引发的免疫应答及肺部出现的病理改变与温度呈负相关。By studying the effect of temperature on the expression of co-stimulatory molecules and lung tissue injury in a murine asthmatic model to explore the correlation of temperature with the severity of allergic diseases, the mice were randomly divided into high temperature group and low temperature group, each group consisted a model group and a control group. The model groups were sensitized on the 1st and 8th day by abdominal injection of sensitizing solution ( including ling OVA and 4rag AI (OH)3), then inhaled OVA for 7 days begining form day 15. All mice were sacrificed at 18 hours after last OVA stimulation. The control groups were treated with same volume of normal saline. The expression of co-stimulatory molecules in the spleen and peripheral blood cells were detected by Flow cytometry (FCM). Blood IgE, IFN-γ and IL 4 were detected by ELISA meth- od. Lung tissue sections were prepared for histopatbologic examination. The results showed that expression of co stimulatory molecules in the spleen and peripheral blood cells of the asthmatic model mice was significantly increased, especially in the low temperature group which was significantly higher than those in the high temperature group. In addition, the levels of both IgE and IL-4 in the blood of asthmatic mice were significantly increased, with the low temperature group significantly higher than those in the high temperature group. However, level of IFN- γin the blood of the asthmatic mice significantly reduced, and thereduction in the low temperature group was more significantly than in the high temperature group. The inflammation in the lung of the model mice was more serious in the low temperature group than that in the high temperature group. Taken togeth- er, our results suggest that asthma-induced immune response and pathological changes of lungs are negatively correlated with temperature.
