肝细胞癌中TBC1D4表达及其诱导的血清抗体免疫应答

摘　　要：检测肝细胞癌患者肿瘤组织中TBC1D4的蛋白表达及其诱导的自身体液免疫应答,为肝细胞癌患者进行特异性免疫治疗提供部分依据。原核表达,镍鳌合亲和白纯化TBC1D4蛋白分子,免疫组化检测26例肝细胞癌患者癌组织的TBC1D4表达;ELISA方法测定96肝细胞癌患者与150例健康对照者血清中抗TBC1D4蛋白抗体的水平。表达纯化了重组蛋白TBC1D4,并诱导出多克隆抗体。26例癌组织中,9例(34.6%,9/26)TBC1D4蛋白表达水平增高;96例患者血清中,17例(17.7%,17/96)血清抗TBC1D4蛋白抗体阳性,150例健康对照者血清全部为阴性。患者组抗体阳性率显著高于健康对照组(P<0.01)。结论认为,TBC1D4蛋白分子在肝细胞癌组织中表达增高,并具有免疫原性,可能是肝细胞癌免疫治疗的一个良好的靶抗原。To investigate TBC1D4 as a potential candidate target antigen in hepatocellular carcinoma therapy, the TBC1D4 pro- tein was expressed in prokaryotic system and purified with nickel chelate affinity chromatography column. Immunohistochemi cal method was used to detect the expression of TBC1D4 in hepatocellular carcinoma tissues. Specific antibodies against TBC1D4 in sera were also detected by ELISA method. The TBC1D4 protein was successfully expressed and purified. Specific polyclonal antibody was obtained after the immunization. 9 （34.6 %, 9/26） out of the 26 hepatocellular carcinoma tissue sam- ples were TBC1D4 positive. The prevalence of TBC1D4 antibodies in patients with hepatocellular carcinoma were significantly higher than those in healthy donors （17.7% vs. 0%, P〈0.01）. TBC1D4 protein was expressed and induced spontaneous anti- body responses in hepatocellular carcinoma patients. TBC1D4 might be a valuable candidate antigen in hepatocellular carcinoma immunotherapy.
