摘  要:目的对沙枣总酚的辐射防护作用进行研究。方法采用不同剂量X射线照射人皮肤成纤维细胞(Human Skin Fibroblasts,HSF),构建辐射损伤模型,以MTT法检测不同剂量沙枣总酚对HSF增殖的影响。结果 X射线对HSF的增殖抑制作用与辐射剂量呈正相关。与2、4、8 Gy辐射组比较,16 Gy辐射组增殖抑制作用有显著差异(P<0.01)。采用16 Gy一次性X射线照射构建辐射损伤模型,模型组与不同剂量沙枣总酚组在2,24,48 h(除100μg/ml-48 h组以外)时段均有显著统计学差异(P<0.01)。照射后2 h,辐射防护作用呈现剂量依赖性,但各剂量给药组的辐射防护作用均随辐射后时间延长而减弱。结论 X射线辐射对HSF具有明显损伤作用,而沙枣总酚对其具有一定的保护作用。Objective To screen the anti - radiation activity of total polyphenols from Elaeagnus angustifolia seeds. Methods The radiation injury model of Human skin fibroblasts (HSF) were established by irradiating HSF with different dosages of X - ray, then MTT assay was used to detect the cell's proliferation activity after being treated by different concentrations of total polyphenols. Results The proliferation inhibition effect of X - ray on HSF was positively correlated with the irradiated dose. This kind of effect in 16 Gy group was more significant compared with 2 Gy,4 Gy and 8 Gy groups (P 〈 0.01 ) respectively. Compared with model group,after the cell was irradiated by 16 Gy X- ray,every total polyphenols group was statistically significant at all- time points (P 〈 0.01 ) except 100 μg/ml -48 h. After irradiation for 2h, radiation protection effect became dose dependent, but the radiation protection effect of the dosage groups decreased with the elongation of time. Condusion X - ray irradiation can induce obvious damage to HSF. Total polyphenols can protect HSF from radiation.
