Runt相关转录因子3基因甲基化在胃癌组织中的表达及临床意义

摘　　要：目的 探讨胃镜活检组织Runt相关转录因子3（RUNX3）基因甲基化在胃癌中的临床意义.方法 研究对象选自武汉协和医院2010年1月至2012年1月60例病理确诊的原发性胃癌患者,其中男36例,女24例,年龄（55.6±10.1）岁；另选60例同期门诊健康者作为正常对照组,其中男35例,女25例,年龄（55.2±9.8）岁.用甲基化特异性聚合酶链反应（MSP）检测胃癌组织及正常对照组织中RUNX3基因启动子甲基化状态.结果采用SPSS 11.0统计软件分析.结果 胃癌组织RUNX3基因启动子甲基化率（61.7％,37/60）显著高于正常对照组织（3.3％,2/60,P＜0.05）.RUNX3基因启动子甲基化率在性别、年龄、发病部位和幽门螺杆菌（HP）感染中的差异无统计学意义（P＞0.05）；在Lauren＇s分型、组织学分级、肿瘤最大径、淋巴结转移、远处转移和TNM分期中的差异有统计学意义（P＜0.05）.结论 胃镜活检织RUNX3基因启动子甲基化与组织学分型、肿瘤大小、有无转移及TNM分期有关.Objective To investigate the clinical significance of Runt-related transcription factor 3 （RUNX3） gene methylation in gastroscope biopsy specimens of patients with gastric cancer.Methods Sixty patients with primary gastric cancer （36 males and 24 females） at the age of （55.6 ± 10.1） years admitted to our hospital from January 2010 to January 2012 were selected as experimental group,and 60 healthy volunteers （35 males and 25 females） at the age of （55.2 ± 9.8） during the same period served as control group.Methylation specific polymerase chain reaction （MSP） was used to detect the methylation status of Runx3 gene promoter in the gastroscope biopsy specimens of 60 gastric cancer patients and normal controls.The results were analyzed by using SPSS software.Results The methylation ratio of Runx3 gene promoter was significantly higher in gastric cancer patients （61.7％,37/60） than in normal controls （3.3％,2/60,P ＜ 0.05）.The Runx3 gene promoter methylation ratio was was related with gender,age,location and HP infection （P ＞ 0.05）,but with Lauren＇s classification,histological grade,tumor size,lymph node metastasis,distant metastasis and TNM staging （P ＜ 0.05）.Conclusion Runx3 gene promoter methylation in gastroscope biopsy specimens may be related with histological classification,tumor size,metastasis and TNM staging.
