护骨胶囊对成骨细胞增殖、分化影响

摘　　要：目的：探讨护骨胶囊对体外培养的大鼠成骨细胞增殖、分化的影响。方法：采用多次酶消化法分离出大鼠颅盖骨的成骨细胞,采用MTT法和碱性磷酸酶法分别检测护骨胶囊对成骨细胞增殖和分化的作用;采用SYBRgreenI嵌合的RT-PCR方法检测Runx2mRNA表达水平。结果：护骨胶囊在浓度0.025~2.5μg/mL对成骨细胞有促增殖的作用,护骨胶囊在浓度0.025~25μg/mL对碱性磷酸酶的活性均有提高的作用,护骨胶囊在浓度25μg/mL下,对Runx2mRNA水平有上调的作用。结论：护骨胶囊对成骨细胞有促增殖、促分化的作用,可能与上调Runx2mRNA水平有关。Objective ： To investigate the effect of Hugu Capsule on proliferation and differentiation in primary osteoblasts. Methods ： Bone cells were obtained by enzyme digestion from the segregated neonatal SD rat skull.The effects of Hugu Capsule on the proliferation and differentiation of rat osteoblasts were evaluated by the MTT method, measuring the activity of alkaline phosphatase, respeetively.Runx2 expression was analyzed by real-time fluorescent quantitative reverse transcriptase polymerase chain reaction （ qRTPCR ） using SYBA Green I . Results ： With the concentration of 0.025-2.5 μg/mL, Hugu Capsule had significant effect on the ROB＇s proliferation.The ALP activity was significantly increased at the concentration from 0.025μg/mL to 25 μ g/mL.With the concentration of 25 μ g/mL, Hugu Capsule can increase the Runx2 mRNA level. Conclusions ： Hugu Capsule can stimulate the proliferation and differentiation of osteoblasts in vitro, and the reason may be connected with the up regulation of Runx2 mRNA level.
