摘 要:目的探讨以杏仁渣为原料提取杏仁蛋白的最优工艺条件。方法通过设计单因素实验和正交实验,将粉碎的杏仁渣加入磷酸盐溶液中水浴加热搅拌,离心,测定上清液的吸光度,计算提取率,确定提取工艺。结果采用磷酸盐法提取杏仁蛋白的工艺为,温度50℃、料液比1∶30,提取1.0 h,盐浓度为0.01 mol/L,蛋白质提取率为72%。结论采用磷酸盐法提取杏仁蛋白,对蛋白没有破坏,提取率较高,为其进一步开发奠定了基础。Objective To extract almond protein by phosphate method from among dregs,and confirm the best process conditions. Methods Based on designing and analyzing single factor experiment and orthogonal experiment, the dregs and phosphate solution were mixed, heated in water bath and stin'ed, centrifugally separated, absorbance of liquid supernatant was measured, and extraction rate was calculated to confirm process parameter. Results The process conditions of extracting almond protein by phosphate method is temperature 50℃, feed liquid ratio 1:30,time 1.0 h, salting strength 0.01 mol/L, and protein extract rate is 72%. Conclusions Extraction of almond protein by phosphate method will not destroy protein and is high in extraction rate.
