摘 要:目的观察吗啡预处理对大鼠大脑中动脉缺血再灌注损伤后大脑组织形态学改变、梗死面积及神经功能的影响。方法将雄性Wistar大鼠随机分为假手术组、模型组及吗啡组,每组12只。假手术组不阻断脑血流,模型组和吗啡组按LONGA方法阻断大脑中动脉2h后再开放灌注。模型组和吗啡组在脑缺血前60min分别腹腔注射生理盐水及1mg/kg吗啡2mL。再灌注24h时,测定大鼠神经功能缺损评分,苏木精-伊红染色观察缺血脑组织形态学改变,红四氮唑(TTC)染色计算脑梗死面积。结果假手术组大鼠神经功能及脑组织形态学正常,大脑组织无梗死。与模型组比较,吗啡组脑组织形态学病理改变较轻,脑梗死面积减少(t=11.22,P<0.05),神经功能缺损评分降低(t=5.86,P<0.05)。结论吗啡预处理能减轻大鼠局灶脑缺血再灌注损伤,改善神经功能。Objective To observe the effect of morphine preconditioning on brain histomorphology, infarct size and neu- rological function after middle cerebral artery occlusion in rats. Methods Thirty-six male Wistar rats were evenly randomized to sham operation group, model group and morphine group. Cerebral blood flow of rats in the sham operation group was not blocked. The middle cerebral artery of rats in the model group and morphine group was blocked for 2 hours as described by L0nga, and then opened again for reperfusion. The rats in model group and morphine group were intraperitoneally injected with normal saline 2 mL and morphine 1 mg/kg, respectively, 60 min before ischemia, and reperfusion for 24 h. Neurologic impairment was scored. Mor- phologic changes of ischemic brain tissue were observed after HE staining, and infarct size of the brain calculated with TTC stai- ning. Results Nerve function and brain histomorphology of rats in the sham operation group were normal. Compared with those in the model group, the pathologic changes of brain tissue was milder and area with cerebral infarction was smaller (t = 11.22 ,P〈 0.05) and neurological impairment score was lower (t=5.86,P〈0.05) in rats of the morphine group. Conclusion Morphine preconditioning can relieve focal cerebral ischemia and improve nerve function.
