摘  要:目的:观察大鼠脑出血(ICH)后不同时间点血肿周围组织Rho激酶Ⅱ(ROCKⅡ)表达及其与凝血酶的关系。方法:180只健康雄性SD大鼠随机分为假手术组(S组)、ICH组和阿加曲班组(ICH+A组)。ICH组和ICH+A组采用自体血注入法建立大鼠ICH模型,ICH+A组于造模后颅内原位注入阿加曲班注射液(0.5 mg/L);S组在相同位点2次注入等容量0.9%氯化钠液。免疫组织化学法观察ICH后不同时间点血肿周围组织ROCKⅡ表达情况及对凝血酶表达的影响。蛋白质印迹(West-ern blotting)法检测ICH组血肿周围ROCKⅡ蛋白表达。并采用Longa和Forelimb Use Asymmetry神经功能缺损评分法对ICH48 h后S组、ICH组、ICH+A组进行神经功能评定。结果:ICH诱导血肿周围ROCKⅡ表达显著增加。造模后0.5 h,ICH组和ICH+A组血肿周围可见ROCKⅡ阳性细胞表达,24 h达高峰,以后逐渐下降,2组表达趋势类似,但在12 h后(包括12 h)相应时间点ICH+A组的ROCKⅡ表达水平显著低于ICH组(P<0.01)。S组病灶周围ROCKⅡ的表达水平均显著低于ICH组和ICH+A组,且细胞着色浅,并且无变化趋势。ICH组Western blotting蛋白条带所显示ROCKⅡ蛋白表达趋势和免疫组织化学结果一致。Longa和Forelimb Use Asymmetry神经功能缺损评分法均显示,ICH后48 h大鼠神经功能缺损评分ICH组和ICH+A组较S组显著增加(P<0.01),而ICH+A组显著低于ICH组(P<0.01)。结论:ROCKⅡ参与ICH后脑损伤,凝血酶可能通过促进ROCKⅡ的表达而产生ICH后脑损伤作用。Objective:To investigate the expression of Rho kinase Ⅱ(ROCKⅡ) around hematoma in rats after intracerebral hemorrhage(ICH) at different time points and its relationship with thrombin.Methods:A total of 180 healthy male SD rats were randomly divided into sham group(S group),ICH group and argatroban group(ICH+A group).ICH model was established by injecting autologous blood.Argatroban was injected in situ at a dose of 0.5 mg/L.The expression of ROCKⅡ around hematoma at different time points and its effect on thrombin were detected by immunohistochemical method.The expression of ROCKⅡ protein around hematoma was detected by Western blotting.Nerve functions of rats were assessed at 48 h by Longa score and forelimb use asymmetry test.Results: Expression of ROCKⅡ around hematoma was found at 0.5 h and reached to a maximal value at 24 h in ICH group and ICH+A group.Subsequently it decreased gradually.However,at the corresponding time points after 12 h,the expression levels of ROCKⅡ in ICH+A group were obviously lower than those in ICH group(P0.01).The expression levels of ROCKⅡ in S group were obviously lower than those in ICH group and ICH+A group,and the trend has not changed.The results of ICH group by Western blotting method and immunohistochemical method were consis-tent.Neurological deficit scores in ICH group and ICH+A group at 48 h increased significantly than those in S group(P0.01),while neurological deficit scores in ICH+A group were significantly lower than those in ICH group(P0.01).Conclusions: ROCKⅡ play a role in brain injury after ICH.Thrombin may have a role in brain injury after ICH through promoting the expression of ROCKⅡ.
