喉鳞癌中microRNA-197表达水平的研究

摘　　要：目的:检测喉鳞癌细胞株Hep-2中microRNAs(miRNAs)的表达谱,并测定喉鳞癌组织中miRNA-197(miR-197)的表达水平。方法:采用miRNAs芯片技术检测喉鳞癌细胞株Hep-2、正常人支气管上皮细胞株HBEAs-2B中miRNAs的表达,得到miRNAs表达谱。根据miRNAs芯片结果,挑选Hep-2中表达显著上调的miR-197,并采用实时定量逆转录聚合酶链反应(realtime quantitative reverse-transcriptase polymerase chain reaction,RT-PCR)验证miR-197在20例喉鳞癌组织及10例癌旁正常组织中的表达。结果:miRNAs芯片结果表明,Hep-2细胞株中有166个miRNAs上调,如miR-197、miR-133b等;47个miRNAs下调,如miR-519d、miR-20b等。RT-PCR检测结果显示,miR-197在喉鳞癌组织中表达水平显著高于癌旁正常组织(P<0.05)。结论:miR-197在喉鳞癌中表达显著上调,可能作为喉鳞癌的特异性miRNA在喉鳞癌的发生发展过程中有重要作用。Objective：To explore the expression profiling of microRNAs（miRNAs）in human laryngeal squamous carcinoma cell line Hep-2 and to investigate the expression level of miRNA-197（miR-197）in human laryngeal squamous carcinoma.Methods：The expression profiling of miRNAs in laryngeal squamous carcinoma cell line Hep-2,and bronchial epithelial cell line HBEAs-2B was detected by using miRNAs chip.Real-time quantitative reverse-transcriptase polymerase chain reaction（RTPCR）was used to quantify the expression levels of miRNA-197（miR-197）in 20 laryngeal squamous carcinoma tissues and 10 normal paracancerous tissues.Results：The results of miRNAs chip showed that 166 miRNAs were significantly up-regulated in Hep-2,such as miR-197,miR-133b,etc.And 47 miRNAs were down-regulated in Hep-2,such as miR-519d,miR-20b,etc.The expression level of miR-197 was significantly higher in laryngeal squamous carcinoma samples compared with that in normal paracancerous tissues（P〈0.05）.Conclusions：Expression of miR-197is up-regulated in laryngeal squamous carcinoma and it may play a role as a specific miRNA in laryngeal squamous carcinoma.
