摘  要:目的研究茶多酚对苯并(a)芘诱导的人单核细胞热休克蛋白(HSP70)和生长分化因子-15(GDF-15)表达的影响。方法健康人单核细胞,按完全随机实验设计分成4个组:①空白对照组;②溶剂对照组[(1‰二甲基亚砜(DMSO),3‰S9];③苯并(a)芘染毒组,以不同浓度苯并(a)芘(0.1、0.5、1、5、10μmol/L)染毒24h;④茶多酚预处理组,以50μg/mL的茶多酚预处理6h后,再以不同浓度苯并(a)芘(0.1、0.5、1、5、10μmol/L)染毒24h,分别应用蛋白印迹法和酶联免疫吸附试验(ELISA)法研究茶多酚对苯并(a)芘诱导的单核细胞HSP70和GDF-15表达情况的影响。结果苯并(a)芘抑制了单核细胞HSP70表达,同时促进GDF-15的分泌;茶多酚预处理组单核细胞HSP70表达与对照组相比差异无统计学意义(P<0.05)。茶多酚预处理组GDF-15分泌没有显著性变化。结论茶多酚可以提高HSP70表达,从而通过内源性的保护机制抵制苯并(a)芘对单核细胞造成的损伤,同时细胞本身通过提高GDF-15表达,降低苯并(a)芘对单核细胞造成的损伤。Objective To investigate the effects of tea polyphenols on HSP70 and GDF-15 expression in monocyte induced by benzo(a)pyrene[B(a)P]. Methods Human monocytes were randomized into 4 groups:( Ⅰ ) blank control group ( Ⅱ ) solvent control group; (Ⅲ ) B(a) P group exposed to B(a) P at the concentration of 0.1, 0.5, 1, 5 and 10 μmol/L, in culture medium for 24 hours: (Ⅳ) cells were pretreated with TP for 6 hours before treated with B(a)P as above in TP-pretreated groups, Western blot was used to measure the expression of HSP70 and the supernatants were collected and analyzed by ELISA to quantify the release of GDF-15 from the cells. Results B(a)P inhibited the expression of HSP70 in TP-pretreated groups and GDF-15 secretion increased with B (a)P increasing. Compared with the control group, there was statistical significance ( P d0.05) in a dose-effect relationship. There was not significant difference in HSP70 expression levels among TP-pretreated groups and control group. Conclusion It is suggested that TP could improve HSP70 expression, which activate the endoge- nous protective mechanism against B(a)P damage to the monocytes, meanwhile, by increasing GDF-15 expression to reduce B(a)P damage for cells.
