腹部开放伤海水浸泡导致大鼠胃黏膜损伤的实验研究

摘　　要：目的观察腹部开放伤合并海水浸泡大鼠胃黏膜破坏,炎性反应和细胞凋亡的变化。方法 40只雄性Wistar大鼠随机分为4组:即正常对照组(A);腹部开放伤组(B);腹部开放伤加淡水浸泡组(C);腹腔开放伤加海水浸泡组(D),每组10只动物。浸泡1 h后处死全部动物,计算胃黏膜溃疡指数;组织学损伤积分;测定胃黏膜组织TNF-α;IL-6含量;免疫组化检测Caspase-3蛋白;TUNEL法检测凋亡细胞。结果 B组、C组和D组大鼠胃黏膜溃疡指数、组织学损伤积分、炎症介质水平、Caspase-3蛋白表达和凋亡细胞百分比均比A组明显升高(P﹤0.01);但是D组胃黏膜溃疡指数、组织学损伤积分、TNF-α、IL-6水平、Caspase-3蛋白表达和凋亡细胞百分比与B组、C组大鼠比较亦有明显增高(P﹤0.05)。结论腹腔开放伤海水浸泡大鼠胃黏膜破坏较为明显,其机制可能与胃黏膜炎性介质的释放以及细胞凋亡密切相关。OBJECTIVE To investigate the gastric mucosal damage, inflammation and apoptosis in rats with abdominal wound and seawater immersion. METHODS 40 male Wistar rats were randomly divided into 4 groups （n = 10, each group） ： A ： normal control group ; B ： abdominal wound group ; C ： abdominal wound and freshwater immersion group; D ： abdominal wound and seawater immersion group. All animals were sacrificed after lh immersion. Calculated gastric ulcer in- dex, gastric histological score; measured the gastric mucosal levels of TNF-ctand IL-6, the expression of Caspase-3 and apoptosis index. RESULTS The results of gastric ulcer index, in expression, the percentages of apoptotic cells in group B, C and D rats were significantly higher than those in group A （P 〈 0.01） ; but the gastric ulcer index, gastric histological score, levels of inflammatory mediators, Caspase-3 protein expression, the percentage of apoptotic cells in the group D were significantly higher than group B and group C （P 〈 0.05 ）. CONCLUSION The gastric mucosal lesion in rats with abdominal wound and seawater immersion is more obvious, the release of inflammatory mediators and apoptosis in gastric mucosa may have close relationship with the gastric mucosal lesion.
