IL-1β诱导肺癌A549细胞CEA过表达对其浸润迁移的影响

摘　　要：目的探讨IL-1β对肺腺癌A549细胞CEA表达的影响及IL-1β诱导的CEA对肺癌A549细胞迁移侵袭能力的影响。方法体外培养的A549细胞分别经IL-1β50、100和200 ng/ml刺激24 h,Western-blot和免疫细胞化学方法检验CEA蛋白的表达;采用细胞Transwell实验和侵袭实验检测IL-1β诱导的CEA对肺癌A549细胞迁移和侵袭能力的影响。结果不同浓度的IL-1β(50、100、200 ng/ml)处理A549细胞后,CEA的表达量较空白对照组增高,且呈剂量依耐性(P<0.05);Transwell试验中对照组、100 ng/ml IL-1β组和加兔抗人CEA多抗组穿过Transwell小室膜的细胞数分别为(52.8±5.5)、(117.2±12.2)和(83.6±5.2)个/视野,侵袭试验中对照组、100 ng/ml IL-1β组和加兔抗人CEA多抗组穿过Matrigel胶的细胞数分别为(37.4±6.1)、(92.6±6.7)和(47.8±6.4)个/视野,均为100 ng/ml IL-1β组高于对照组(P<0.05);加兔抗人CEA多抗组低于100 ng/ml IL-1β组(P<0.05)。结论 IL-1β可能通过诱导CEA过表达,促进肺癌A549细胞的迁移和侵袭。Objective To observe the influence of IL-1β on the expression of carcinoembryonic antigen （CEA） in human lung cancer cell line A549 and of the CEA induction on the migration and invasion of A549. Methods A549 cells cultured in vitro were pretreated with different concentrations of IL-1β （50,100 and 200 ng/ml）for 24h, and CEA expression was detected by Western- blot and inmmnocytochemical assay. Transwell assay and invasion assay were used to test the influence of CEA in- duced by IL-1β （100ng/ml） on the migration and invasion of A549 cell line. Results The CEA expressions in A549 cells pretreated with IL-1β at concentrations of 50,100 and 200 ng/ml were significantly higher than that in the control cells, and the expression increased in a dc,se-dependent manner （P〈0.05）. In the Transwell assay, the numbers of cells through the mem- brane of Transwell chambers in the control group, 100ng/ml IL-1β group and CEA polyclonal antibody group were respectively （52.8 ± 5.5）, （117.2 ± 12.2）and （83.6 ±5.2）/view. In the invasion assay, the numbers of the cells through the Matrigel in Transwell chambers in the control group, 100ng/ml IL-1β group and CEA polyclonal antibody group were respectively（37.4 ±6.1）, （92.6± 6.7）and （47.8 ± 6.4）/view. For both assays, the cell numbers in 100ng/ml IL-1β group were significant higher than those in the control group and in the CEA polyelonal antibody group （ P 〈 0.05）. Conclusions IL-1β may promo e the migation and invasion of human lung cancer cell line A549 by inducing CEA over- expression.
