抗原负载的DC及CIK对结肠癌细胞SW1116杀伤活性的比较研究

摘　　要：目的探讨树突状细胞(dendritic cells,DCs)以及CIK(cytokine induced cells)细胞对结肠癌细胞SW1116的体外杀伤作用,为结肠癌的治疗提供新的治疗手段。方法用IL-4、GM-CSF、TNFα等细胞因子诱导培养DC细胞,并用抗原进行负载;用CD3Ab、IFNγ、IL-2、IL-1α等诱导培养CIK细胞,分别用抗原负载的DC、CIK细胞对人结肠癌细胞SW1116进行体外杀伤活性实验,比较两者对SW1116的杀伤效果。结果抗原负载的DC和CIK对SW1116均有较强的杀伤效果,分别达到64.10%和67.70%。结论抗原负载的DC及CIK对人结肠癌细胞SW1116在体外具有较强的杀伤作用。Objective to investigate the killing effect of dendritic cells and CIK （cytokine induced cells） on colon cancer cell SWl116 in vitro ,provide a new therapeutic method for the treatment of colon cancer. Methods The method is that DC cells are induced and cultured using cell factor,including IL-4, GM-CSF, TNFaet al. ,CIK cells are induced and cultured using cell factor, including CD3Ab, IFNy, IL -2, IL-1αet al. DC cells and CIK cells are further loaded by specific antigens. The loaded DC and CIK cells are then respectively used to do colon cancer SW1116 cells in vitro killing experiment and compare their killing effect. Final results show that both antigen loaded DC and CIK cells have strong killing ability to SWl116 cancer cells. Results The relative killing effects are 64.10% and 67.70%. Conclusion There are stronger killing effects of DC and CIK cells loaded by antigen on Colorectal cancer cell S W1116 in vitro.
