摘 要:目的观察莪术提取物(莪术醇)对人脑恶性胶质瘤CHG-5细胞的生长抑制作用,探讨莪术提取物诱导人脑恶性胶质瘤细胞凋亡、影响细胞增殖的机制。方法应用人工培养CHG-5细胞,配制不同质量浓度的莪术提取物作用于该细胞;经过一定时间后,利用倒置显微镜观察CHG-5细胞的生长状态及形态学变化情况;采用MTT法检测莪术提取物对CHG-5细胞的生长抑制作用;丫啶橙染色观察药物作用下对细胞形态的改变及对凋亡的影响;采用Annexin V-FITC/PI双染色法检测凋亡,而细胞周期进程的变化及凋亡率则采用流式细胞仪技术检测。根据实验及统计分析结果分析莪术提取物与其原癌细胞株CHG-5之间的关系及临床意义。结果莪术醇对人脑恶性胶质瘤CHG-5细胞增殖表现抑制效应,显示为浓度依赖性与时间依赖性(P均<0.05);莪术醇的浓度可以影响人脑恶性胶质瘤CHG-5细胞密度、形态;莪术醇可以增加人脑恶性胶质瘤CHG-5细胞的凋亡,显示出浓度依赖性(P<0.05)。结论莪术提取物对CHG-5细胞的生长具有抑制作用,其强度与药物浓度和作用时间呈正相关。莪术提取物对CHG-5细胞的凋亡具有诱导作用,随着时间和剂量的增加,凋亡率也随之增加。莪术提取物影响CHG-5细胞的周期分布,于G1/S期阻滞细胞周期的进程,这可能是莪术提取物影响肿瘤的发生及发展机制。Objective It is to observe the inhibition of zedoary extract on growth of human malignant gliomas CHG - 5 cells, and approach the mechanisms of zedoary extract in inducing human brain malignant glioma cell apoptosis, affecting cell proliferation. Methods Artificial culture CHG - 5 cells were treated by different concentrations of zedoary extract. After a cer- tain time, the growth state of the CHG - 5 cells and morphological changes were observed by inverted microscope; the growth inhibition of zedoary extract on CHG -5 cell was detected by MTT assay; cell morphology changes and apoptosis were observed by acridine orange staining; apoptosis was detected by Annexin V -FITC/PI double staining, but changes in cell cycle pro- gression and apoptosis rate were detected by flow cytometry. The relationship of zedoary extract and original cancer cell line CHG - 5 and its' clinical significance were analyzed according to the results of experimental and statistical analysis. Results The inhibitory effect of curcumol was performed in human brain malignant glioma CHG - 5 cell proliferation, and with concen- tration-dependent and time-dependent manner (P 〈 0.05 ) ; the density and form of human brain malignant glioma CHG - 5 cell were affected by concentration of curcumol ; the apoptosis of human malignant gliomas CHG - 5 was increased by curcumol with concentration-dependent (P 〈 0.05). Conclusion Zedoary extract can inhibit the growth of CHG -5 cell; its strength was positively correlated with drug concentration and action time. Zcdoary extract can induce apoptosis of CHG - 5, the apop- tosis rate was increased with the increasing of time and dose. Zedoary extract can affect CHG - 5 cell cycle, and arrest cell cy- cle progression at GI/S phase, which may be the mechanism of zedoary extract in affecting the occurrence and development of tumors.
