摘 要:目的研究HIF-1、MGMT、CD133在人低级别胶质瘤及胶质母细胞瘤细胞中的表达差异及三者在胶质瘤母细胞瘤中表达的相关性。方法收集自2010年12月至2012年12月手术切除病理证实的胶质母细胞瘤(WHOⅣ级)33例作为实验组,同期收集低级别胶质瘤25例(WHOI-II级)作为对照组,收集脑深部肿瘤皮层造瘘正常脑组织5例作为阴性对照组。检测不同组别组织中HIF-1、MGMT、CD133的表达,应用SPSS 13.0进行分析。相关统计分析采用Spearman等级相关分析。结果在正常脑组织中HIF-1、MGMT、CD133不表达,在低级别胶质瘤组和胶质母细胞瘤组中其阳性表达率差异具有统计学意义(P<0.05)。通过RT-PCR法分别测定HIF-1、MGMT、CD133在正常脑组织、低级别胶质瘤及胶质母细胞瘤中mRNA的相对定量表达,进行相关性分析,三者均存在正相关(HIF-1与MGMT r=0.91,P<0.05;HIF-1与CD133 r=0.85,P<0.05;MGMT与CD133 r=0.80,P<0.05)。结论 HIF-1、MGMT、CD133阳性表达率随肿瘤级别增高呈显著升高,在胶质母细胞瘤中其表达呈正相关,提示三者在胶质母细胞瘤的发生、发展过程中存在协同作用,为胶质母细胞瘤的治疗提供新的方向。Objective To study the expression of HIF-1, MGMT and CD133 in low grade gliomas and glioblasto- mas and to investigate the correlation among HIF-1, MGMT and CD133 in glioblastomas . Methods All sample tissues were collected from surgeries and pathologically confirmed in Hebei Tumor Hospital between Dec 2010 and Dee 2012. Thirty-three samples of glioblastomas were used as experimental group and 25 samples of low grade gliomas as control group. Five normal cortical tissues from the deep brain tumors were used as negative control group. The mRNA and pro- tein levels of HIF-1, MGMT and CD133 were examined in various grade glima and normal brain tissue. SPSS13.0 soft- ware was used for statistical analysis and differences with a probability value of P〈0.05 were considered statistically sig- nificant and ~=0.05 was considered as size of the test . Results There was no expression of HIF-1, MGMT and CD133 in normal brain tissue. In contrast, the expression levels of HIF-1, MGMT and CD133 were statistically significant differ- ence between low grade gliomas and glioblastoma (P〈0.05). The correlation analyses showed that there were significant positive correlations among the HIF-1, MGMT and CD133 expression (rHIF-1 and MGMT =0.91, P〈0.05; rHIF-1 and CD133=0.85, P〈0.05; rMGMT and CD133 =0.80, P〈0.05 ). Conclusions The expression levels of HIF-1, MGMT and CD133 are significantly ele- vated with the increase of tumor grade. There are significant positive correlations of expression levels between HIF-I and MGMT, between HIF-1 and CD133, and between MGMT and CD133, indicating a coordinated effects among among H1F-ltx, MGMT and CD133 in the development, recurrence of glioblastoma. The present study provides a new direction for glioblastoma treatment.
