摘  要:目的 探讨Twist蛋白在肾肿瘤组织中的表达及与几种多药耐药蛋白表达之间的关系.方法 应用免疫组化SABC技术检测人肾癌组织不同区域的Twist蛋白、多药耐药相关蛋白(multidrug resistance-associated protein,MRP)、P糖蛋白(P-gp)、肺耐药蛋白(lung resistance-related protein,LRP)、拓扑异构酶Ⅱ(topoisomerase Ⅱ,TOPOⅡ)的表达.结果 在肿瘤边界区,Twist蛋白、MRP、P-gp、LRP、TOPOⅡ均为阳性表达;而在肿瘤中心区,Twist蛋白和P-gp的表达为阴性,MRP、LRP、TOPOⅡ蛋白仍为阳性表达,但MRP和TOPOⅡ在肿瘤中心区表达强于肿瘤周边区(P〈0.01),而LRP在肿瘤中心区的表达弱于肿瘤周边区(P〈0.01).结论 Twist基因的表达与肾肿瘤转移、浸润以及耐药性有关.Objective To investigate the relationship between the expressions of Twist protein in renal carcinoma tissue and several drug resistance proteins. Methods The expressions of Twist protein, multidrug resistance - associated protein (MRP), P- glycoprotein (P- gp), lung resistance protein (LRP), and topoisomerase Ⅱ (TOPO Ⅱ) in different zones of human renal carcinoma tissues were detected using streptavidin - biotin - peroxidase complex (SABC) immune histochemistry technique. Results Immunostaining results showed that the expressions of Twist protein, MRP, P - gp, LRP, and TOPO Ⅱ were positive in the boundary zone of renal carcinoma, while in the cancer center zone, the expressions of Twist protein and P - gp were negative. MRP, LRP, and TOPO Ⅱ were still positively expressed in cancer center zone, and the expression levels of MRP and TOPO Ⅱ were higher in cancer center zone than those in boundary zone (P 〈0.01 ), while the expression level of LRP was lower than that in boundary- zone (P 〈0. 01 ). Conclusion Twist gene is related to metastasis, infiltration, and drug resistance in renal carcinoma.
