摘 要:目的将大黄素制成固体分散体以提高大黄素的生物利用度,研究大黄素固体分散体对大鼠酒精性肝损伤的保护作用。方法以聚乙烯吡咯烷酮为载体制备大黄素固体分散体。SD大鼠给予酒脂乳剂制备酒精性肝损伤模型,分别给予大黄素和大黄素固体分散体,连续给药8周。测定总胆固醇(TC)、甘油三酯(TG)、丙氨酸氨基转移酶(AST)、天门冬氨酸氨基转移酶(ALT)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)。结果大黄素固体分散体能显著降低酒精性肝损伤大鼠血清和肝组织中TG和TC含量,降低血清和肝组织中AST和ALT活性;降低肝组织中MDA含量,提高血清和肝组织中SOD和GSH活力。结论大黄素固体分散体对酒精性肝损伤大鼠具有保护作用。OBJECTIVE Solid dispersion of emodin was prepared to enhance the bioavailability of emodin, and the protective effects of emodin solid dispersion on alcohol-induced liver damage in rats were investigated. METHODS Polyvinylpyrrolidone was used as the vehicle for the preparation of emodin solid dispersion. Sprague Dawley (SD) rats were fed an alcohol emulsion to create an alcohol-induced liver damage model. The rats were then treated with emodin and emodin solid dispersion for 8 consecutive weeks. Levels of total cholesterol (TC), triglyceride (TG), alanine aminotransferase (AST), aspartate aminotransferase (ALT), malondialdehyde (MDA), glutathione (GSH-Px), and superoxide dismutase (SOD) in the rats were determined. RESULTS Emodin solid dispersion significantly reduced TG and TC levels, as well as AST and ALT activities, in serum and liver tissues of rats with alcohol-induced liver damage. In addition, MDA content in liver tissues was lowered, and SOD and GSH activities in serum and liver tissues were improved. CONCLUSION Emodin solid dispersion has protective effects against alcohol-induced liver damage in rats.
