摘 要:目的探讨Exendin-4对肿瘤坏死因子α(TNF-α)诱导下的大鼠肾小球系膜细胞氧化应激和转化生长因子β1(TGF-β1)表达的影响。方法培养大鼠肾小球系膜细胞,分为对照(A)组、TNF-α(B)组、TNF-α+Exendin-4(C)组。培养48 h后,使用分光光度比色法测定细胞上清液超氧化物歧化酶(SOD)活性、丙二醛(MDA)、谷胱甘肽(GSH)的含量,采用流式细胞术检测ROS含量,ELISA法检测TGF-β1蛋白表达。结果 B组系膜细胞培养上清SOD(15.46±1.19)U/ml、GSH(18.48±1.16)μmol/ml,均低于A组(P<0.01)。B组细胞培养上清MDA(4.56±0.26)nmol/ml、TGF-β1(375±14)pg/ml,ROS平均荧光强度(MFI)为(1100±40),与A组比较差异有统计学意义(P<0.01),Exendin-4干预可拮抗TNF-α刺激下系膜细胞的上述改变。结论 Exendin-4可抑制TNF-α诱导的系膜细胞氧化应激和TGF-β1表达,提示Exendin-4可能会对大鼠有保护作用。Objective Objective To investigate the effect of Exendin-4 on the oxidative stress and expression of TGF-β1 in cultured glomerular mesangial cells induced by TNF-α in vitro. Methods The cultured mesangial cells were divided into control group, TNF-α group and TNF-α+ exendin-4 group. After 48h, the supernatant was collected. The activity of SOD, the content of MDA, GSH, and ROS, and the expression of TGF-β1 were detected by spectrophotometry, flow cytometry and ELISA respectively. Results Compared with the control group, the activity of SOD (15.46 ± 1.19)U/ml and the content of GSH (18. 48± 1.16)μmol/ml in the TNF-α group was significantly decreased, while the content of MDA (4. 56± 0. 26) μmol/ml and ROS of mean fluorescence intenity (1100 ± 40), and the expression of TGF-131 (375 ± 14)pg/ml were increased, the differences were statistically significant (P〈0.01). Exendin-4 could inhibit the changes of the mesangial cells (P〈0.01). Conclusion Exendin-4 can reverse the effect of TNF-α on the oxygen stress and expression of TGF-β1 in mesangial cells. The reasonable application of exendin-4 may be helpful in preventing and treating diabetic nephropathy.
