摘  要:目的建立多波长HPLC法同时测定养血清脑颗粒中没食子酸,原儿茶酸,绿原酸,咖啡酸,芍药苷,芍药内酯苷和阿魏酸的量。方法采用Aglient Zorbax SB-C18色谱柱(4.6 mm×250 mm,5μm),以0.08%磷酸水溶液-乙腈为流动相,梯度洗脱,体积流量0.95 mL/min,检测波长为280 nm(0~17 min),320 nm(17~25 min),230 nm(25~33 min),320 nm(33~43 min),柱温30℃。结果没食子酸,原儿茶酸,绿原酸,咖啡酸,芍药苷,芍药内酯苷,阿魏酸的线性范围分别为0.942~9.42μg/mL,0.408~4.08μg/mL,1.01~10.1μg/mL,0.332~3.32μg/mL,3.24~32.4μg/mL,7.34~73.4μg/mL,0.607~6.07μg/mL(r≥0.999 8);平均回收率(n=6)均在95%~105%范围内。结论所建立的HPLC法简便,灵敏,准确,可用于养血清脑颗粒质量控制。AIM To establish an HPLC method for simultaneous determination of gallic acid,protocatechuic acid,chlorogenic acid,caffeic acid,albiflorin,paeoniflorin and ferulic acid in Yangxue Qingnao Granules.METHODS The chromatographic method was carried out on an Aglient Zorbax SB-C18 column(4.6 mm × 250 mm,5 μm) with acetonitrile-0.08% phosphoric acid solution as the mobile phase in gradient elution mode.The flow rate was 0.95 mL/min.The detection wavelength was set at 280 nm(0-17 min),320 nm(17-25 min),230 nm(25-33 min),320 nm(33-43 min).The column temperature was 30 ℃. RESULTS The linear ranges of gallic acid,protocatechuic acid,chlorogenic acid,caffeic acid,albiflorin,paeoniflorin and ferulic acid were 0.942-9.42 μg/mL,0.408-4.08 μg/mL,1.01-10.1 μg/mL,0.332-3.32 μg/mL,3.24-32.4 μg/mL,7.34-73.4 μg/mL and 0.607-6.07 μg/mL(r≥0.999 8),respectively.The average recoveries(n=6) were between 95% and 105%.CONCLUSION The method is simple,sensible,accurate and can be used for the quality control of Yangxue Qingnao Granules.
