摘  要:目的探讨川芎嗪(tetramethylpyrazine,TMP)对重组人表皮生长因子(recombinant human epidermal growth factor,rhEGF)诱导的牛晶状体上皮细胞(bovine lens epithelial cell,BLEC)增殖的抑制作用。方法采用BLEC进行体外原代及传代培养,利用rhEGF诱导BLEC增殖,同时用低、中、高三种不同浓度的TMP作用处于增殖状态下的BLEC。采用MTS法测定细胞增殖情况。通过RT-PCR方法检测正常对照组、增殖对照组和低、中、高浓度TMP组的Bcl-2的基因表达水平。结果 MTS检测发现,TMP以剂量依赖方式抑制rhEGF诱导的BLEC增殖。RT-PCR发现增殖对照组的Bcl-2表达增高,TMP可以下调Bcl-2的表达水平,两者差异有统计学意义。结论 TMP能有效抑制rhEGF诱导的BLEC增殖。Objective To detect the inhibitory effects of tetramethylpyrazine (TMP) on proliferation of bovine lens epithelial cells (BLEC) induced by recombinant human epidermal growth factor (rhEGF). Methods BLEC were primari- ly cultured and sub-cultured. BLEC which have been induced to proliferate by rhEGF were incubated with low, middle and high concentration of TMP. The activity of BLEC was detected by MTS, Bcl-2 mRNA expression was detected in the normal control group, control group and low, middle, high concentration of TMP groups by RT-PCR. Results Proliferation of BLEC induced by rhEGF was inhibited by TMP with dose-effect manner. The expression of Bcl-2 mRNA was significantly increased in control group, and the expression of Bcl-2 was reduced in groups of low, middle and high concentration of TMP. Conclusions TMP can effectively inhibit the proliferation of BLEC induced by rhEGF.
