摘  要:目的分析脂肪干细胞以及自体富血小板血浆作为载体复合物在体内进行血管化组织工程脂肪构建的可行性以及应用效果。方法取吸脂术后健康成人的脂肪组织,并分离出脂肪干细胞,进行原代以及传代培养,以BrdU标记第3代脂肪干细胞,并进行定向诱导,配置细胞悬液,取细胞悬液5mL+富血小板浆工作液100μL(实验组,n=10)或DMEM培养基100μL(对照组,n=10)+纤维蛋白胶0.5mL,分别移植于裸鼠的背部皮下。结果人脂肪干细胞的形态表现较好.免疫荧光阳性标记率在90%以上;实验组新生组织湿重为(0.1525±0.0142)g,脂肪组织微血管数为(45.9±5.4)支,对照组分别为(0.0858±0.0104)g、(18.2±3.7)支,实验组均显著高于对照组,差异有统计学意义(P〈0.05);两组新生组织经免疫荧光染色显示。细胞和以及部分微血管的内皮细胞核均呈现出绿色荧光。结论脂肪干细胞、自体富血小板血浆以及纤维蛋白胶载体在机体内能够促进形成血管化组织工程脂肪,且自体富血小板血浆能够促进组织工程构建物重建血运.从而提高种子细胞的成活率。Objective To analyze the feasibility and application effect of construction of autologous adipose stem ceils and platelet-rich plasma carrier complex in vivo by tissue engineering. Methods Adipose stem cells were isolated from healthy adults after liposuction, conducted primary culture and subculture. The 3rd generation adipose stem cells were labeled by BrdU and orientation induced. Ceil suspension 5 mL+platelet-rich plasma working fluid 100 μL (experiment group, n = 10) or DMEM medium 100 μL (control group, n = 10) +fibrin glue 0.5 mL were taken and transplanted subcutaneously in nude mice. Results The morphology of human adipose stem cells morphologic expression were better, immunofluorescence positive mark rate was 90% or more; experiment group of new tissue wet mass was (0.1525±0.0142) g, fat tissue microvascular number was (45.9±5.4) branch, the control group were (0.0858±0.0104) g, (18.2±3.7) branch respectively, the experiment group were higher than control group, the differences were statistically significant (P 〈 0.05); ceils and microvascular endothelial cells showed the green fluorescence by immunofluorescence staining. Conclusion Adipose stem cells, autologous platelet-rich plasma and fibrin glue carrier in the body can promote the formation of vascularized adipose tissue engineering and autologous platelet-rich plasma can promote tissue engineering constructs revascularization, thereby it can easily improve the survival rate of seed cells.
