摘  要:目的:探讨PI3K/Akt信号通路在外源性硫化氢诱导人卵巢癌细胞凋亡中的作用。方法:用硫化氢供体硫氢化钠(10-5、10-4和10-3mol/L)处理卵巢癌细胞株SKOV3细胞12 h、24 h和48 h后,采用MTT法测算细胞增殖抑制率、流式细胞术检测细胞凋亡,采用实时荧光定量PCR和Western blot分别检测p-Akt mRNA和蛋白的表达,观察PI3K/Akt信号通路激动剂胰岛素样生长因子-1(IGF-1)对外源性硫化氢作用的影响。结果:10-4和10-3mol/L的硫氢化钠处理24 h和48 h,以浓度和剂量依赖的方式增加了细胞增殖抑制率和细胞凋亡率,下调了p-Akt的表达(P<0.05)。IGF-1取消了硫氢化钠诱导的SKOV3细胞增殖抑制和凋亡增加(P<0.05)。结论:外源性硫化氢通过PI3K/Akt信号通路诱导人卵巢癌细胞凋亡,抑制增殖。[ Abstract] Objective: To investigate the role of PI3 K/Akt pathway in the apoptosis and proliferation induced by exogenous hydro- gen sulfide (H2S) in human ovarian carcinoma cells. Methods: The human ovarian carcinoma cells were treated with H2S donor sodium bi- sulfide (NariS) (10-5 tool/L, 10-4 mol/L and 10-3 tool/L) for 12, 24 and 48 hours. MTY assay was used to detect the inhibition of cell proliferation. The apoptosis of cells were measured by flow cytometry. The mRNA and protein expressions of p - Akt in human ovarian carcino- ma cells were measured by Real - time PCR and Western blot. PI3K/Akt pathway agonist insulin - like growth factor 1 ( IGF - 1 ) was used in this study. Results: The proliferations were significantly decreased and the rates of apoptosis were significantly increased in human ovarian carcinoma cells by treatment with sodium bisulfide ( 10 -4 mol/L and 10 -3 mol/L) for 24 and 48 hours in a dose - dependent and time - de- pendent manner. The expression of p - Akt was down - regulated by treatment with sodium bisulfide ( 10 -4 mol/L and 10 -3 mol/L) for 48 hours and sodium bisulfide ( 10 -3 tool/L) for 24 and 48 hours in a dose - dependent and time - dependent manner. The increase of apoptosis and inhibition of proliferation induced by sodium bisulfide were reversed by PI3K/Akt pathway agonist IGF - 1. Conclusion: Exogenous hy- drogen sulfide inhibits the proliferation and promotes the apoptosis in human ovarian carcinoma cells through PI3K/Akt pathway.
