CD_8T淋巴细胞在大鼠缺血再灌注脑组织中表达的变化

摘　　要：目的观察大鼠大脑中动脉闭塞(middle cerebral artery occlusion,MCAO)脑缺血模型再灌注不同时间点的脑组织中CD8T淋巴细胞表达的动态变化,探讨CD8T淋巴细胞与缺血性脑损伤的关系。方法选择成年雄性SD大鼠35只,随机分为假手术组,缺血再灌注24h、3d、5d、7d、10d和14d组,每组5只。采用Zea-Longa线栓法制作大鼠MCAO脑缺血模型,HE染色显示脑缺血再灌注组织病理学形态,SP法免疫组织化学染色显示CD8分子表达的变化,光镜下计数CD8T淋巴细胞数。结果再灌注7d和10d组,脑组织大量神经元细胞质、胞核界限模糊、核固缩溶解,细胞间出现大小不等的空泡。CD8T淋巴细胞于再灌注24h组即开始表达,再灌注7、10d组达到高峰,随后下降。再灌注各时间点组CD8T淋巴细胞数显著高于假手术组(P<0.01)。CD8T淋巴细胞大量表达于缺血再灌注脑组织,且主要分布于脑梗死边缘区。结论 CD8T淋巴细胞参与了脑缺血再灌注损伤。Objective To study the relation between CD8T lymphocytes and ischemic brain injury by observing the expression of CD8T lymphocytes in rat ischemia/reperfusion （I/R） tissue at dif- ferent time points in a rat middle cerebral artery occlusion （MCAO） model. Methods Thirty-five adult male SD rats were randomly divided into sham operation group,24-h I/R group , 3- day I/ R group, 5-day I/R group, 7-day I/R group, 10-day I/R group and 14-day I/R group （5 in each group）. A rat MCAO model was established by Zea-Longa occlusion. Pathological morpholo- gy of I/R tissue was observed with HE staining and expression of CD8 molecules was detected with SP immunohistochemical staining. The number of CD8T lymphocytes was calculated under light microscope. Results Maasive neuron cytoplasm,blurred nucleus line,pyknotic dissolution in brain tissue, and different size vacuoles between nerve cells were found in 7-day I/R group and 10--day I/R group. The CD8T lymphocytes bagan to express 24 h after I/R injury and reached its peak on days 7 and 10 after I/R injury and then decreased. The number of positive CD8T lympho- cytes was significantly greater in different I/R groups than in sham operation group （P〈0.01）. The CD8T lymphocytes were mainly expressed in I/R brain tissue and distributed in marginal zone of ischemic stroke. Conclusion CD8T lymphocytes are involved in cerebral I/R injury.
