摘 要:目的:观察祛风通窍方对VD大鼠血液流变学及海马CA1区神经元细胞变化影响。方法:SD大鼠90只,假手术组12只,余78只经2次Morris水迷宫筛选符合要求的VD大鼠60只,随机分为模型组、尼莫地平组(西药组)、祛风通窍方高、中、低剂量组,每组12只。双侧颈总动脉永久性结扎法(2-VO)制备VD模型。流变快测仪检测血液流变学指标,电镜观察海马神经元细胞变化。结果:与假手术组比较,模型组大鼠血液流变学、神经元细胞变化、细胞凋亡指数(AI)明显升高(P<0.05);西药组、祛风通窍方高、中剂量组神经元细胞超微结构有效改善,AI及血液流变学指标与模型组比较均显著降低,差异有统计学意义(P<0.05)。结论:祛风通窍方能改善血液流变学,有效减轻VD大鼠神经元损伤,抑制神经元细胞凋亡。Objective: To observe the effect of Qufeng Tongqiao Decoction on hemorheology and hippocampal CA1 neuron changes of VD rats. Methods: A total of 90 SD rats,12 rats were randomly assigned to sham group;after screen by Morris Water Maze test, the other 60 qualified rats were randomly divided into model group, nimodipine group(N group), high, medium and low dose Qufeng Tongqiao decoction groups (Q groups) 12 each. Permanent bilateral carotid artery ligation (2-VO) was used to VD model. The hemorheology changes were detected by quick rheometer, and the hippoeampal neurons by electron microscopy. Results: Compared with sham group, blood rheology, neuronal changes, and apoptosis index (AI) in model group were significantly higher (P 〈 0.05); compared with model group of the uhrastrueture neuron of in N group and medium and high dose Q groups was significanly improved, while AI and blood rheology were significantly lower; the difference was statistically significant (P 〈 0.05). Conclusion: Qufeng Tongqiao decoction can improve the blood rheology, effectively reduce the neuron injury in VD rats, and inhibit neuronal apoptosis.
