摘 要:目的探讨NEP/Nogo-66 1-40peptides(NEP1-40)对视网膜缺血再灌注损伤的保护作用。方法SD大鼠分成正常对照组,视网膜缺血再灌注组和NEP1-40组。采用单眼生理盐水前房加压60min再灌注24h制作视网膜缺血再灌注损伤模型,石蜡切片,HE染色,光学显微镜下观察视网膜细胞形态结构的变化,免疫组化观察各组视网膜中DNA修复酶Ku70的表达变化,PCR检测各组视网膜中Ku70 mRNA表达变化。结果缺血再灌注损伤组(IR组)视网膜结构紊乱,细胞肿胀变性,Ku70蛋白及mRNA表达较正常组下降(<0.01),NEP1-40组视网膜结构明显改善,Ku70蛋白及mRNA表达较IR组升高(<0.01)。结论 NEP1-40对大鼠视网膜缺血再灌注损伤的保护作用可能与增加Ku70蛋白及mRNA的表达相关。Objective To study the protective effect of NEP/Nogo-66 1-40 peptides(NEP1-40) on retinal ischemia-reperfusion injury and its possible mechanism. Methods The SD rats were divided into normal control, retinal ischemia-reperfusion(IR) and NEP1-40 groups. The IR injury model of SD rats was established by pressure injection of normal saline into monocular anterior chamber for 60 minutes and reperfusion for 24h. HE staining was used to observe the retinal cell morphology, the expression of DNA repair enzyme KuT0 protein and mRNA was studied with the immunohistochemistry and PCR assay respectively. Results The expression levels of Ku70 protein and mRNA in retina were decreased in IR group than in the normal control(P 〈 0.01), with the cell swelling and degeneration in IR group. However, the expression levels of Ku70 protein and mRNA in retina were increased in NEP1-40 group than in the IR group (P 〈 0.01), with the improvement of cell injury in NEP1-40 group. Conclusion The protective effect of NEP1-40 on retinal isehemia-reperfusion injury might be related to the upregulation of Ku70 expression.
