摘 要:目的探讨化疗药物、细胞因子诱导的杀伤细胞(CIK细胞)、特异性细胞毒性T淋巴细胞(CTL)对乳腺癌MCF-7细胞CD55high亚群的杀伤效果。方法用不同血药浓度的化疗药物多西他赛、表阿霉素和氟尿嘧啶杀伤乳腺癌MCF-7细胞;抽取人外周血诱导培养CIK细胞,杀伤CD55high细胞和CD55low细胞;术中取患者腋下淋巴结体外诱导培养肿瘤特异性CTL,检测CTL对CD55high细胞和CD55low细胞体外杀伤活性,分析化疗药物、CIK细胞、特异性CTL对CD55high亚群细胞的杀伤效果。结果多西他赛、表阿霉素和氟尿嘧啶对CD55high亚群细胞杀伤率明显低于其对MCF-7细胞杀伤率;CIK细胞CD55high亚群细胞杀伤率(42.72±4.36)%;特异性CTL对CD55high、CD55low和MCF-7细胞杀伤率分别为(52.86±4.45)%、(22.41±2.83)%、(21.67±4.15)%,CD55high组明显高于CD55low和MCF-7组细胞的杀伤率(P<0.01)。特异性CTL组与CIK细胞组和多西他赛(0.4μg/mL)组3组比较,有显著性差异(P<0.01)。结论特异性CTL对CD55high细胞具有特异性杀伤效果。Objective To investigate the activities of cytokine induced killer cells (CIK cells) and specific cytotoxic T lymphocytes (CTL) in killing CD55high subgroup in MCF-7 breast cancer cell line. Methods The killing effects of CIK cells, and specific CTLs in diminishing CD55Q subgroup cells were analyzed and compared with those of three chemotherapy drugs. First, the MCF-7 breast cancer cells were treated with different concentrations of three chemotherapeutic drugs, docetaxel, epirubicin and fluorouracil. Then immune cells were isolated from human peripheral blood and were induced to become CIK cells. They were then used to treat CD55"~" and CD55~w subpopulations in MCF-? cells. Finally, lymphocytes were isolated from surgical specimens of human axially lymph nodes, and were induced to become tumor-specific CTL in vitro. Their activities in killing CD55"~Qh and CD55~~w subpopulations in MCF-7 cells were measured as well. Results The killing rates of docetaxel, epirubicin and fluorouracil on CD55hiQ" subgroup MCF-? cells were much lower than on total MCF-7 cells. The killing rate of CIK cells on CD55high subgroup cells was(42.72 +4.36)%. The killing rates of specific CTLs on CD55high, CD55=low, and total MCF-7 cells were(52.86 + 4.45 ) %, (22.41 +2.83 ) %, and (21.67 + 4.15 ) %, respectively. Conclusion Specific CTL possessed more effective killing effects on CD55high cells.
