摘 要:目的 研究白介素-1(IL-1)预刺激对反义受体相关激酶-2寡核苷酸(IRAK-2ODN)抑制IL-1激活核因子-kappa B(NF-κB)的影响。方法 实验分IL-1预处理组和非预处理组。以脂质体介导反义IRKA-2ODN转染293细胞,以夹心酶联免疫吸附测定法(ELISA)检测NF-κB的含量。结果 IL-1非预处理组反义IRAK-2ODN不能抑制NF-κB活性。Objective To investigate the effects of IL-1-pretreatment on the inhibition of IL-1-induced NF-κB activation by antisense IL-1 kinase-2 receptor- associated (IRAK-2) oligonucleotide. Methods Antisense IRAK-2 oligonucleotide mediated by lipofectin encapsulation was used to transfect 293 cells. The levels of nuclear factor-kappa B(NF-κB) were determined by sandwich enzyme-linked immunosorbent assay. Results Antisense IRAK-2 oligonucleotide did not inhibit IL-1-induced NF-κB activation without IL-1-pretreatment. However, NF-κB activation was significantly inhibited by antisense IRAK-2 oligonucleotide in a time- (5~24 h) and dose- (1~8 (g) dependent manner if the cells were pretreated with IL-1. Conclusion IL-1-pretreatment is necessary for inhibition of IL-1-stimulated NF-κB activation by antisense IRAK-2 oligonucleotide.
