多发性骨髓瘤患者血清IL-8水平及其基因启动子多态性研究

摘　　要：目的研究IL-8基因多态性对血清IL-8水平的影响以及其在多发性骨髓瘤中的作用。方法检测正常人和多发性骨髓瘤患者血清中IL-8的水平,并应用PCR-限制性片段长度多态性(PCR-RFLP)技术研究IL-8-251 a/T位点多态性。结果多发性骨髓瘤患者和正常对照组之间IL-8-251 A/T位点多态性各基因型频率和等位基因频率相比有显著性差异(P均<0.05)。多发性骨髓瘤患者不同基因型血清中的IL-8水平均高于正常对照组IL-8水平,其中基因型为A/A的患者产生IL-8量最少,基因型为T/T的患者产生IL-8因子最多,而基因型为T/A的患者IL-8量居中(P<0.05)。结论 IL-8-251 A/T基因多态性与多发性骨髓瘤患者血清中IL-8水平有密切关系,预示着IL-8-251 A/T多态性可能参与多发性骨髓瘤患者疾病网络调控。Objective It is to observe the influence of IL- 8 gene polymorphism on the levels of serum IL- 8 in multiple myeloma patients, and approach its＇ role. Methods Polymorphism of IL - 8 gene was identified with polymerase chain reaction- restriction fragmented length polymorphism （PCR - RFLP） in 50 multiple myeloma patients and 50 healthy people, the serum IL - 8 levels were measured by enzyme - linked immunosorbent assay （ ELISA）. Results There was significant difference in fre quencies of allele and genotype in IL- 8 -251A/T polymorphism between patients and controls （P 〈0.05 ）. The serum IL- 8 levels of multiple patients were significantly higher than that of healthy controls, besides, the IL - 8 levels in patients with different genotypes were as follows ： T/T 〉 T/A 〉 A/A. Conclusion IL - 8 - 251 A/T gene polymorphism is related to the lev els of IL - 8 in patients with multiple myeloma. IL - 8 gene polymorphism may be involved in the mechanism of regulation net work in multiple myeloma patients.
