健脾疏肝方对CCl_4诱导小鼠肝纤维化抑制作用的实验研究  被引量:3

摘  要:目的:观察中药复方健脾疏肝方对CCl4诱导小鼠肝纤维化的抑制作用,并探寻其作用机制。方法:将80只雄性清洁级C57BL/6J小鼠随机均分为4组,即正常组、模型组、健脾疏肝组及活血化瘀组。除正常组外,其余3组腹腔注射20%CCl4橄榄油剂1 mL/kg(2次/周,共8周)。正常组腹腔注射等量100%橄榄油剂(2次/周,共8周)。第5周起健脾疏肝组及活血化瘀组分别给予健脾疏肝方和扶正化瘀胶囊灌胃(1次/d,共4周)。实验结束后检测小鼠肝组织基质金属蛋白酶(MMP-1)及其抑制物(TIMP-1)、血清透明质酸(HA)、层黏连蛋白(LN)、三型前胶原(PCⅢ)及四型胶原(Ⅳ-C)水平,采用光镜观察小鼠肝脏病理组织学改变。结果:健脾疏肝组、活血化瘀组TIMP-1、HA、LN、PCⅢ、Ⅳ-C水平均显著低于模型组(P<0.01);健脾疏肝组TIMP-1、HA、LN、PCⅢ、Ⅳ-C水平均显著低于活血化瘀组(P<0.01);与空白组相比,其余3组MMP-1均明显升高(P<0.01),但彼此无明显差异(P>0.05)。结论:健脾疏肝方对CCl4诱导的小鼠肝纤维化具有良好的治疗作用。Objective:To observe the effect of Jianpi Shugan Formula (JPSGF) on hepatic fibrosis induced by CC14 in mice compared with Huoxue Huayu Formula (HXHYF), and to explore its mechanism. Methods:80 male clean grade C57BL/6J mice were randomly divided into 4 groups : the normal group, the model group, the JPSG group and the HXHY group. Except the normal group, other 3 groups were abdominally injected with 20% CC14 olive oil solution a dose of 1 mL/kg body weight twice a week for eight weeks. And the normal group was abdominally injected with 100% olive oil of the same dose twice a week for eight we^ks. Since the fifth week, the JPSG group and the HXHY group were intragastricly administered JPSGF and HXHYF respectively with a dose of 0.2 mL/10 g once a day for 4 weeks. The normal and the model groups were given the same volume of deionized water. After the end of experiment, the levels of liver tissue matrix metalloproteinases ( MMP - 1 ) , its inhibitor ( TIMP - 1 ) , serum hyalu- ronic acid ( HA ) , laminin protein ( LN), procollagen Ⅲ ( PC Ⅲ ) and collagen IV ( 1V - C ) were detected bv Enzvme- linked im-munosorbent assay. Meanwhile, histological changes in mice liver pathology were observed by light microscopy. Results : The levels of TIMP - 1, HA, LN, PC Ⅲ, IV - C of the JPSGF and HXHYF groups were all significantly lower than those of the model group ( P 〈 0. 05 ). The levels of TIMP - 1, HA, LN, PC Ⅲ, IV - C of the JPSGF group were significantly lower than those of the HXHYF group (P 〈 0. 05 ). Compared with the normal group, the MMP - 1 levels of other three groups were all significantly higher (P 〈 0. 05 ). However, the MMP - 1 levels of the three groups had no significant difference ( P 〈 O. 05 ). Conclusion : JPSGF shows a striking effect of inhibition and treatment of hepatic fibrosis in mice caused by CC14.
