摘 要:目的:研究川芎嗪对辐射致血虚证小鼠骨髓细胞IL-6和IL-18基因表达的影响,为阐明川芎嗪补血和免疫调节作用的分子机制提供依据。方法:采用3.5 Gy60Coγ射线全身1次性照射制备小鼠血虚证模型;外周血白细胞计数;计算胸腺指数和脾指数;RT-PCR检测骨髓IL-6和IL-18 mRNA水平。结果:川芎嗪显著升高放射线致血虚证小鼠外周血白细胞数量(P<0.05)和脾指数(P<0.01),上调骨髓细胞IL-6(P<0.05)和IL-18(P<0.01)基因表达,对胸腺指数无显著影响。结论:川芎嗪促进骨髓细胞IL-6和IL-18基因的表达可能是其升血和调节免疫功能的机制之一。Objective:To study the effect of ligustrazine on IL -6 and IL - 18 gene expression in bone marrow cell of blood de- ficiency mice induced by radiation and provide the basis for clarifying the molecular mechanism of blood enriching and immuno- regulatory function of ligustrazine. Methods :The blood deficiency mice were induced by using a single dose of 3.5 Gy 60 Co γ radi- ation. Peripheral blood leukocytes were counted and the thymus and spleen indexes were calculated. IL -6 and IL - 18 mRNA in bone marrow were measured by RT - PCR. Results : Ligustrazine can significantly raise the peripheral blood leukocytes number ( P 〈0.05 ) and the spleen index (P 〈 0.01 ) of blood deficiency mice induced by radiation and up- regulate IL- 6 (P 〈 0.05 ) and IL - 18 (P 〈0.01 ) gene expressions of bone marrow cell of the mice,but had no significant effect on thymus index of the mice. Conclusion:Ligustrazine can promote IL- 6 and IL- 18 gene expression in bone marrow cells, which is probably one of the mechanisms of blood proliferation and immunoregulatory function of the ligustrazine.
