摘 要:目的探讨14-3-3蛋白各亚型在人脑胶质瘤中表达及其与肿瘤病理分级的关系。方法采用免疫组化ABC法检测14-3-3蛋白各亚型在80例人脑胶质瘤和10例正常脑组织标本中的表达。结果在正常脑组织中14-3-3蛋白7个亚型主要表达于神经元,但是ε、ζ和θ亚型在胶质细胞中弱表达。大部分胶质瘤组织中β、ε、ζ、η和θ亚型均有表达,且它们的免疫反应评分随着肿瘤的级别增高而增高(P<0.05)。结论β、ε、ζ、η和θ在人脑胶质瘤的发生中可能起着重要的作用,尤其是β和η亚型。Purpose To explore the correlation between the expression of 14-3-3 isoforms and the pathological grades in gliomas. Methods Ten normal brain tissues and eighty cases of gliomas were enrolled in this study. 14-3-3 isoforms were detected by immuno- histochemical avidin-biotin-peroxidase complex method. The relationship between the expression of 14-3-3 isoforms and the pathological grades in gliomas was analyzed. Results In the normal brain, 14-3-3 isoforms immunoreactivity was localized mainly in the neuronal somata and processes, while only weak expression of ε, ζ and θ was found in some glial cells. However, β、ε、ζ、η and θ isoforms' immunoreactivity was seen in the majority of astrocyte tumor samples and its immunoreactivity score was increased markedly with an in- crease in the pathologic grade of human astroeyte tumors. Conclusion 14-3-3 isoforms, especially β and η, may play an important role in the tumorigenesis of human astrocyte tumors.
