摘 要:目的探讨miR-26b过表达在人脂肪细胞分化过程中对各种脂因子分泌的影响。方法以miR-26b过表达病毒感染人脂肪前体细胞并诱导分化;收集不同时间点的细胞培养基上清,采用酶联免疫吸附法(ELISA)检测各个时间点的脂因子(白介素-6、瘦素、抵抗素、肿瘤坏死因子-α)分泌量。结果与对照组比较,在miR-26b过表达病毒感染的人脂肪前体细胞分化过程中,白介素-6、瘦素分泌量降低,而抵抗素分泌量升高;两组均未检测到肿瘤坏死因子-α。结论 miR-26b在一定程度上可以改善人脂肪细胞炎症状态及改善胰岛素抵抗,为肥胖治疗提供潜在靶点。Objective To investigate the influence of overexpression of miR-26b on the secretion of adipokines dur- ing human adipocyte differentiation. Methods Human preadipocytes were infected with the hsa-miR-26b over-expressing lentivirus and were induced to differentiate, and then the levels of adipokines (IL-6, leptin, resistin, TNF-α) at different time points during differentiation were measured by ELISA. Results Compared with control group, decreased secretions of both IL-6 and leptin, and increased secretion of resistin were found during the differentiation of human adipocytes in miR- 26b overexpressed group. However, the secretion of TNF-a was not measured in both groups. Conclusion The miR-26b can improve the inflammation and insulin resistance of human adipocytes, which will provide potential targets for obesity treatment.
