摘 要:【目的】探索棉花黄萎菌黑色素合成途径中的相关基因。【方法】通过设计多对引物,对新疆棉花黄萎菌强致病力菌株V592进行了聚酮合成酶基因(PKS)的PCR、克隆和测序,并对所测序列进行基因结构及进化分析。【结果】获得真菌DHN黑色素合成途径中的一个关键酶—聚酮合成酶基因(PKS)的全序列,为6 742bp(登录号:KC422576)。序列分析显示,该基因由4个外显子和3个内含子组成,其开放阅读框(OR f)编码2 189个氨基酸;具有I型聚酮合成酶基因(PKS I)的结构特征。以KS编码氨基酸序列构建系统关系树,结果表明产生相同多聚酮次生代谢物质的真菌具有相同的进化来源,V592与产生黑色素(melanin)的其他真菌处于相同的进化分支上,而产生其他次生代谢产物的真菌则分别形成不同的进化分支。【结论】PKS基因与棉花黄萎菌黑色素的产生有关。[ Objective ] The purpose of this research was to explore the gene involved in melanin blosyn- thesis of Verticillium dahlae from Cotton. [ Method] Polyketide Synthase gene (PKS) was determined from a virulent defoliating isolate V592 of Verticillium dahlae by PCR, cloning and sequencing. [ Result] PKS gene comprised 6742 nucleotides ( GenBank accession No. KC422576) including 4 exons and 3 introns, and ORF encode 2189 amino acid. It showed typical PKSI genetic characteristics. Phylogenetie tree was performed based on alignments of amino acid sequences of KS domain among V592 and other fungi. It showed that these fungi producing secondary metabolite including Polyketide had same phylogenetic relationships. V592 clus- tered together with other fungi producing melanin, while other fungi producing other same Polyketide metabo- lite clustered together, respectively. [ Conclusion ] It was indicated that PKSI was closely related to melanin biosynthesis of V. dahlae from cotton.
