摘 要:目的观察健脾化湿法对脾虚湿困证溃疡性结肠炎(ulcerative colitis,UC)大鼠血清干扰素-γ(interferon-γ,IFN-γ)和白介素-4(interleukin-4,IL-4)含量的影响,探讨健脾化湿法治疗UC的作用机制。方法将40只Wistar大鼠随机分为正常对照组、模型组、阳性对照组、健脾化湿组。采用多因素相结合的方法,模拟中医传统病因"久居湿地,饮食失宜,过于劳倦"复制脾虚湿困证大鼠模型,采用2,4,6-三硝基苯磺酸灌肠法复制UC模型。采用酶联免疫吸附测定法检测各组大鼠血清IFN-γ和IL-4含量。结果与模型组比较,健脾化湿组大鼠血清IFN-γ含量显著降低(P<0.05),IL-4含量显著升高(P<0.01),IFN-γ/IL-4比值显著降低(P<0.01)。结论健脾化湿法治疗脾虚湿困证UC的作用机制与降低血清IFN-γ含量,升高血清IL-4含量,抑制1型辅助T细胞介导的炎性反应有关。Objective To observe the effects of spleen-strengthening and dampness-removing (SSDR) prescription on serum interferon-γ (IFN-γ) and interleukin-4 (IL-4) levels in rats with ulcerative colitis (UC) and syndrome of dampness stagnancy due to spleen deficiency and to investigate the action mechanism of SSDR prescription in the treatment of UC. Metimds Forty Wistar rats were randomly divided into normal control group, model group, positive control group, and SSDR group. In the model group, positive control group, and SSDR group, a rat model of dampness stagnancy due to spleen deficiency was induced by simulating the traditional Chinese medicine causes: long stay in the wetland, improper di et, and overstrain; then, a rat model of UC was induced by enema of 2,4,6-trinitrobenzenesulfonic acid. Serum IFN-γ and IL-4 levels were measured by enzyme-linked immunosorbent assay. Results Compared with the model group, the SSDR group had significantly decreased serum IFN-γ level (P〈0. 05), significantly increased serum IL4 level (P〈0. 01), and significantly decreased IFN-γ/IL-4 ratio (P〈0. 01). Conclusion The action mechanism of SSDR prescription involves decreasing serum IFN-γ level, increasing serum IL-4 level, and inhibiting Thl-mediated inflammatory response in the treat- ment of UC with syndrome of dampness stagnancy due to spleen deficiency.
