摘  要:目的 探讨建立小鼠肝癌原位模型并观察其生物治疗的效果.方法 选取Balb/c小鼠来源的ML-3肝癌细胞经肝内直接注射接种至肝脏实质内,与经脾注射肝癌细胞相比较,对其成瘤情况进行观察;并予以生物治疗,观察其疗效.结果 肿瘤细胞接种第6天起,经脾脏注射组以及经肝脏注射各组小鼠均不同程度出现体质量减轻、腹水、黄疸以及腹腔可触及包快.在经脾脏注射组小鼠中,5只小鼠中仅有1只小鼠长出肝脏肿块.其余均腹腔种植转移.在经肝脏直接注射小鼠中,15只小鼠中有10只长出肝癌,腹腔中没有种植转移8例.DC肿瘤疫苗联合OX86单抗治疗组小鼠的全身状况要优于疫苗单独治疗组以及PBS对照组.分别选取对照组小鼠以及治疗组小鼠的脾脏细胞以及腹水细胞样本进行流式细胞分析,治疗组脾脏和腹水细胞中MDSCs,即表达CD11b+ LyC6+细胞(2.77%)低于对照组(11.94%)(x2=5.36,P<0.05).结论 经肝脏直接注射肿瘤细胞较经脾脏注射法能更有效诱导肝癌原位模型,且生物治疗对于该肝癌实验模型有一定效果.Objective To establish an orthotropic tumor model for hepatocellular carcinoma in mice and to explore the effect of biotherapy on this tumor model.Methods ML-3 hepatocellular carcinoma cells derived from Balb/c mice were injected into the mice through intrahepatic injection and intra-spleen injection.The tumor formation and the survival of the mice were observed and the effect of tumor vaccine was studied preliminarily.Results The weight reduction,ascites,jaundice and abdominal mass were found on the 6th day of tumor cells injection.Liver tumor was oberserved in 1 mouse by introspleen injection,and implantation metastasis was found in the other 4 mice.Among the 15 mice through intrahepatic injection,liver tumor was found in 10 mice and 8 mice without implantation metastasis.The overall outcome of the mice received DC tumor vaccine combined with OX86 antibody was better than the vaccine alone and PBS control group.Spleen cell and ascites cell samples of control and treatment group were selected and analyzed by flow cytometry.MDSCs cell of the treatment group which expressed CD11b+ LyC6+ (2.77%) was lower than that of control group (11.94%) (x2 =5.36,P < 0.05).Conclusion The orthotropic tumor model for hepatocellular carcinoma in mice was successfully established by intrahepatic injection rather than intra-spleen injection,and tumor biotherapy was to some extent effective to this orthotropic model.
