摘  要:目的观察阿托伐他汀抑制老年大鼠心肌炎性因子白细胞介素-1β(IL-1β)表达的作用与过氧化物酶体增殖物激活型受体γ(PPARγ)信号转导途径之间的关系。方法分离培养24个月龄大鼠的心肌细胞。将培养的老年大鼠心肌细胞分为空白对照组、溶剂对照组、阿托伐他汀组、PPARγ拮抗剂GW9662+阿托伐他汀组。各组分别加入细胞培养液、二甲基亚砜、阿托伐他汀、阿托伐他汀+GW9662处理。用RT-PCR方法检测各组大鼠心肌细胞的IL-1βmRNA表达水平,用Western blot方法检测各组大鼠心肌细胞的IL-1β蛋白含量。结果阿托伐他汀组IL-1βmRNA表达水平及IL-1β蛋白含量均低于空白对照组、溶剂对照组(P<0.01);阿托伐他汀+GW9662组的IL-1βmRNA表达水平及IL-1β蛋白含量均高于阿托伐他汀组(P<0.05),但仍显著低于空白对照组、溶剂对照组(P<0.05或P<0.01)。结论阿托伐他汀可通过激活PPARγ信号转导途径来下调IL-1β在衰老心肌的表达。Objective To observe the correlation between effect of Atorvastatin in inhibition of Interleukin-1β (IL-1β) gene expression and signal transduction pathway of peroxisome proliferator activated receptor γ (PPARγ) in myocytes of elderly rats. Methods Myocardial cells of 24-month-old rats were isolated culture. Cultural myocytes were divided into control group, dimethyl sulfoxide (DMSO) group, Atorvastatin group and antagonist GW9662 of PPARγ plus Atorvastatin group, and every group was added into cell culture fluid, DMSO, Atorvastatin and Atorvastatin plus GW9662 respectively. The expressions of |L-1β mRNA in the four groups were detected by reverse transcription polymerase chain reaction (RT-PCR) method, and contents of IL-1β protein in the four groups were detected by western blot method. Resuits The expression level of IL-1β mRNA and content of IL-1β protein in Atorvastatin group were significantly lower than those in control group and DMSO group (P 〈0. 01 ) ; the expression level of IL-1βmRNA and content of IL-Iβ protein in Atorvastatin plus GW9662 group were significantly higher than those in Atorvastatin group ( P 〈 0.05 ), but still significantly lower than those in control group and DMSO group (P 〈 0. 05 or P 〈 0. 01 ). Conclusion Atorvastatin may regulate IL-1β gene expression down in aging myocytes by activating signal transduction pathway of PPARγ.
