摘 要:目的观察Rho激酶2(ROCK2)在癫痫大鼠脑内的表达情况及其抑制剂法舒地尔对癫痫大鼠脑电图的影响。方法 75只SD大鼠随机分入正常对照组、6h组、1d组、3d组、5d组、7d组、空白组、癫痫组和法舒地尔组。腹腔注射氯化锂-匹鲁卡品建立癫痫模型。通过免疫组织化学和Western blot方法,比较各组大鼠颞叶、海马区ROCK2表达的差异;对空白组、癫痫组及法舒地尔组大鼠进行脑电监测,分析脑电图变化。结果在海马组织中,3d组、5d组ROCK2的表达水平较对照组升高,差异有统计学意义(P<0.05);在颞叶脑组织中,3d组、5d组、7d组ROCK2的表达水平较对照组均明显升高,差异有统计学意义(P<0.05)。免疫荧光结果显示正常组大鼠与实验组大鼠的海马组织中ROCK2(绿色)与NeuN(红色)在神经元中共表达;颞叶脑组织中ROCK2(绿色)与GFAP(红色)在星形胶质细胞中共表达。脑电监测结果显示空白组大鼠脑电正常。癫痫组大鼠在给予匹鲁卡品约28min后出现痫样放电波形。与癫痫组大鼠相比,法舒地尔组大鼠出现痫样波形的潜伏时间明显延长,频率明显降慢、幅度明显降低(P<0.05))。结论癫痫发作将上调大鼠颞叶和海马区ROCK2的表达,ROCK2抑制剂法舒地尔具有一定的抗癫痫作用。Objective To study the expression of Rho kinase 2 in brain of rat model of epilepsy at diferent time, and the influence of Rho kinase 2 inhibitor fasudi to EEG of epileptic rats. Methods Seventy--five SD rats were randomly divided into normal group(n= 10), 6h group(n= 10), l d group(n=10), 3d group(n=10), 5d group(n=10), 7d group(n=10) , blank group(n 5), epilepsy group(n =5) and fasudil group(n=5). By intraperitoneal injection of LiC1 -- PILO to established the model of epilepsy. By immunohistochemistry and Western blot method, to compare the expression of ROCK2 in the rat temporal lobe and hippocampus. Monitoring the EEG of the control group, epilepsy group and fasudil group rats, and analysis the difference of the EEG. Results In hippocampus, the level of ROCK2 in 3d and 5d group is higher than the control group, the difference was statistically significant (P〈0. 05). In temporal lobe , 3d.5d and 7d group is higher than the control group, the difference was statis- tically significant (P%0. 05). Immunofluorescence showed that in normal rats and experimental rats hip pocampus ROCK2 (green) and NeuN (red) express together and in temporal lobe the ROCK2 and GFAP also express together. EEG monitoring showed, in the blank group EEG was normal. After given pilocarpine about 28mins epileptiform waveforms were found in the epileptic rats. Epileptiform wave also were found in fasudil group, but latency was prolonged, the frequency significantly slow down, amplitude was significantly lower (P〈0. 05). Conclusion Seizures will increase the expression of ROCK2 in tem- poral lobe and hippocampus of rats. ROCK2 inhibitor fasudil has some antiepileptic effect.
