摘 要:目的:探讨B细胞易位基因1(BTG1)在人肾小管上皮细胞株(HK-2)和肾透明细胞腺癌细胞株(786-O)中的表达及BTG1基因对HK-2和786-O细胞周期、细胞凋亡的影响。方法:采用Western blotting方法检测BTG1蛋白在HK-2、786-O细胞株中的表达,构建BTG1表达质粒转染786-O细胞,合成BTG1干扰RNA转染HK-2细胞,通过流式细胞仪(FACS)观察BTG1对HK-2、786-O细胞周期和细胞凋亡的影响。结果:Western blotting检测显示,BTG1蛋白在HK-2中的表达高于786-O。786-O中高表达BTG1蛋白能够促进其凋亡,增加其G0/G1期比率;HK-2中低表达BTG1蛋白能够抑制其凋亡,减少其G0/G1期比率。结论:BTG1可能通过对细胞凋亡及细胞周期的调控抑制肾细胞癌的生长。Objective:To explore the expression of B-cell translocation gene 1 ( BTG1 ) in normal renal tubule epithelial cell lines (HK-2) and renal clear cell cancer cell lines (786-O) and its influence on cell cycle and apoptosis of HK-2 and 786-O.Methods: The expression of BTG1 in HK-2 and 786-O was detected by Western blotting.The BTG1 expression plasmid was constructed and transfected into 786-O.The interference RNA of BTG1 was designed and transfected into HK-2.The effect of BTG1 on cell cycle and apoptosis of HK-2 and 786-O was observed by flow cytometry (FACS).Results:The expression of BTG1 protein was higher in HK-2 than in 786-O. High expression of BTG 1 would promote cell apoptosis and increase the rate of G 0/G1 phase in 786-O.Low expression of BTG1 would inhibit the cell apoptosis and reduce rate of G 0/G1 phase in HK-2.Conclusion:BTG1 can inhibit the growth of renal cell carcinoma through regulation of cell cycle and apoptosis .
