细胞周期素D_1和磷酸酶基因蛋白在喉鳞状细胞癌中的表达及与临床病理特征的关系

摘　　要：目的探讨喉鳞状细胞癌(LSCC)组织中细胞周期素D1(Cyclin D1)和磷酸酶基因(PTEN)蛋白的表达及其与临床病理特征的关系。方法应用免疫组化方法检测50例LSCC组织、20例不典型增生(AH)、15例声带息肉(VCP)和10例正常喉黏膜(NLM)组织中Cyclin D1和PTEN蛋白表达水平,并结合临床病理特征进行分析。结果在LSCC中Cyclin D1和PTEN表达阳性率分别为62.0%和48.0%。LSCC中Cyclin D1表达阳性率高于AH、VCP和NLM,而PTEN表达阳性率低于AH、VCP和NLM(P<0.05)。Cyclin D1和PTEN表达与LSCC分级程度、临床分期和颈部淋巴结转移相关(P<0.05)。LSCC中Cyclin D1表达与PTEN表达呈显著负相关(r=-0.32,P<0.05)。结论联合检测Cyclin D1和PTEN蛋白的表达对提示LSCC恶性程度、临床进展和预测颈部淋巴结转移具有重要意义。Objective To investigate the expression of Cyclin D~ and PTEN in laryngeal squamous cell carcino- mas （LSCC） and their relationship with clinicopathological characteristics. Methods Expressions of Cyclin D1 and PTEN in 50 patients with LSCC, 20 patients with atypical hyperplasia （AH）, 15 patients with vocal cord polyp （VCP） and 10 patients with normal laryngeal mucosa （NLM） were detected by immunohistochemical method, and were analyzed with clinicopathological characteristics. Results The positive expression rates of Cyclin D~ and PTEN in LSCC were 62.0% and 48.0% respectively. The positive expression rate of Cyclin D~ in LSCC was higher than those in AH, VCP and NLM, ＇but the positive expression rate of PTEN was lower than those in AH, VCP and NLM （ P 〈 0.05 ）. The expres- sions of Cyclin D1 and PTEN were correlated with tumor grade, clinical stage and lymph nodes metastasis of LSCC （P 〈 0.05 ）. The expression of Cyclin D1 was negatively correlated with that of PTEN in LSCC （ r = - 0.32,P 〈 0. 05）. Con- clusion Combined detection of expressions of Cyclin D1 and PTEN protein may be closely related with tumor grade, clin- ical stage, prognosis of lymph node metastasis in LSCC.
