摘 要:目的研究华蟾素注射剂在小鼠体内的最大耐受量以及不同剂量华蟾素对荷人胰腺癌细胞株SW1990裸小鼠的抑瘤作用。方法将华蟾素分为100、75、50、30、10、3 g·kg^(-1)共6个不同剂量组并设对照组(生理盐水组),观察该药对CD1小鼠的最大耐受量;根据最大耐受量的结果将荷人胰腺癌细胞株SW1990皮下移植瘤裸小鼠,之后随机分为对照组与吉西他滨组,以及华蟾素12.5、25、50 g·kg^(-1)剂量组,定期测量肿瘤体积,给药21 d后计算抑瘤率。结果华蟾素毒性反应出现在100、75 g·kg^(-1)两剂量组。华蟾素小剂量组抑瘤率为16.95%,中剂量组为30.51%,大剂量组为18.64%,各给药组的平均瘤体积均小于对照组(P<0.05);其中华蟾素中剂量组的抑瘤率较好。华蟾素各剂量组裸小鼠均生长良好,无明显毒性反应。结论华蟾素在CD1小鼠体内的最大耐受剂量为50 g·kg^(-1),华蟾素给药剂量为25 g·kg^(-1)时对荷人胰腺癌细胞株SW1990裸小鼠抑瘤作用最佳。AIM To study the maximum toxicity dose of Huachansu injection and the anti-tumor effects on xenograft nude mice with pancreatic cancer cell SW1990. METHODS The CD1 mice were divided into 6 groups treated with Huachansu injection at 100, 75,50, 30, 10 and 3 g·kg^-1 dosage levels , and the mice in control group were treat-ed with normal saline . The xenograft nude mice with pancreatic cancer cell SW1990 were divided into 5 groups: control group, gemcitabine group and Huachansu group at 3 different dosage levels( 12.5, 25 and 50 g·kg^-1). The size of tu-mor was measured regularly in three weeks. RESULTS The CD1 mice treated with Huachansu injection at 100, 75 g·kg^-1 dosage level had severe toxicity. Otherwise, the anti-tumor experiment showed that the inhibition rate of tumor size of Huachansu injection at low, middle and high dosage levels (12.5, 25 and 50 g·kg^-1)were 16.95% ,30.51% and 18.64% respectively. There were significant differences between the Huachansu groups and the control group (P 〈 0.05). The inhibition rate of tumor size in middle dosage Huachansu group was better than that in low and high dosage Huachansu groups( P 〈 0. 05). CONCLUSION The 50 g·kg^-1 is the maximum toxicity dose of Huachansu injection . The middle dosage Huachansu group (25 g·kg^-1) has better anti-tumor effects on xenograft nude mice with pancreatic cancer cell SW1990.
