摘 要:目的探讨乳结散治疗乳腺增生病的作用机制。方法选用SD雌性未孕大鼠40只,随机分为4组,各10只,即正常组、模型组、乳结散低剂量组、乳结散高剂量组,乳腺增生模型造模成功后给药组予乳结散低剂量汤药、乳结散高剂量汤药灌胃,正常组及模型组均灌服0.9%氯化钠注射液,共30 d,采集标本,检测各组大鼠血清泌乳素(PRL)、雌二醇(E2)、孕酮(P)含量,乳腺组织形态学和第2对乳头高度变化及雌激素受体(ER)的表达。结果乳结散低剂量组、乳结散高剂量组可降低乳腺增生模型大鼠第2对乳头高度(P<0.05),改善血清PRL、E2、P含量(P<0.05),降低增生乳腺组织ER表达(P<0.05)。显著改善乳腺增生的组织细胞学表现。与模型组比较,乳结散低剂量组和高剂量组大鼠乳腺组织上皮排列基本规则,管腔扩张减轻,腺泡数显著减少,乳腺间质纤维结缔组织增生亦明显减轻。结论乳结散可能通过调整雌孕激素比例及ER的表达来改善乳腺增生。Objective To explore the mechanisms of Rujiesan on the treatment of breast hyperplasia. Methods 40 nonpregnant female SD rats were randomly divided into four groups, normal group, model group, low dose Rujiesan group and high dose Rujiesan group. After the modeling, low dose Rujiesan group and high dose Rujiesan group received intragastric administration of low dose and high doses of Rujiesan decoction. Normal group and model group received 0.9% NaC1 injection by gavage for 30 days. The contents of prolactin( PRL), estradiol( E2 ) and pro- gesterone (P) in serum in each group were detected. Breast tissue morphology, the first two pairs of nipple height vari- ation and estrogen receptor(ER) expression were observed. Results The first two pairs of nipple height was reduced after treatment in low dose Rujiesan group and high dose Rujiesan group(P 〈 0.05 ). The contents of .PRL, E2 and P in serum were improved( P 〈 0.05) and ER expression was lessened (P 〈 0.05 ). The performance of histology and cytology of breast hyperplasia was significantly improved. Rat mammary epithelial arrangement was essentially regular, lumen expansion was reduced, the number of aeini was significantly reduced and mammary gland hyperplasia of interstitial fibrous connective tissue also significantly reduced as compared with those in model group. Conclusion Ru- jiesan can improve breast hyperplasia possible by means of adjusting the ratio of estrogen and progesterone and estro- gen receptor expression.
