摘  要:目的:观察不同浓度ZnSO4对浮游状态变异链球菌生长、产酸及其生物膜形成的影响.方法:将变异链球菌UA159分别接种于含ZnSO40、1、2、5、10 mmol/L的BHI培养液中,37℃、厌氧培养.分别于培养4、8、12、18、24h检测各组培养液的OD值、pH值,并进行CFU(Colony-Forming Units)计数.同时,将变异链球菌分别于上述各浓度ZnSO4共同培养24 h,使之在盖玻片上形成生物膜;采用CUF计数法观察各组生物膜中细菌数,并运用激光共聚焦扫描显微镜(CLSM)观察各组生物膜中活菌数及其厚度.实验数据用t检验法进行统计分析.结果:1~10 mmoL/L不同浓度ZnSO4均能抑制浮游态变异链球菌的生长、产酸;并使其生物膜变薄,活菌数减少,且其作用随ZnSO4浓度的增加而增加,各组间差异均有统计学意义(P<0.05).但是,虽然ZnSO4的浓度为10 mmol/L时能使变异链球菌处于生长停滞状态,却仍有一定的产酸能力,且生物膜内的细菌也未被全部杀死.结论:ZnSO4仅能抑制变异链球菌的生长,无法将其彻底杀灭并阻断其产酸.AIM:To investigate the effect of ZnSO4 on the proliferation,acid production and biofilm formation of streptococci mutans (S.mutans).METHODS:S.mutans were anaerobicly cultured in BHI containing ZnSO4 at 0,1,2,5 and 10mmoL/L,for 4,8,12,18 and 24 h,respectively.The pH of the culture suspensions was measured and colony-forming unit (CFU) of S.mutans in the suspensions was examined by UV spectrophotometer at OD600.S.mutans biofilms were formed by culture on glass cover slips and CFU in the biofilms was examined.The biofilms were observed by confocal laser scanning microscope (CLSM).The percentage of vital bacteria was calculated.Data were assessed by SAS8.02 using Student's test (t test).RESULTS:ZnSO4 at higher concentration showed stronger antibacterial effects on S.mutans in both suspension and biofilms (P < 0.05).10 mmoL/L ZnSO4 nearly completely stopped the proliferation of S.mutans in suspension,but the inhibition of acid production was not complete.The mean thickness of biofilm was reduced significantly,but 10 ~ 20% bacteria were still alive in the biofilm.CONCLUSION:ZnSO4 to S.mutans is bacteriostasis instead of bactericidal.ZnSO4 should be mantained at high concentration to exert effective antibacterial effects.
