摘  要:目的:研究细胞周期通路中的细胞周期蛋白B1(Cyclin B1)在人、大鼠、树鼩不同种属的肝癌、癌旁及正常肝组织中的表达和意义。方法:采用实时荧光定量-PCR(Q-PCR)和蛋白质印迹法检测人(38例肝癌及对应癌旁组织、10例正常肝组织)、大鼠(15例肝癌及其相应癌旁组织、14例正常肝组织)和树鼩(10例肝癌及其相应癌旁组织、10例正常肝组织)中Cyclin B1的mRNA和蛋白表达水平。结果:人肝癌组织Cyclin B1mRNA的表达量(0.011 4±0.009 6)高于正常肝组织(0.004 8±0.002 9),差异有统计学意义,P=0.033;高于癌旁组织Cyclin B1mRNA的表达量(0.003 7±0.002 7),差异有统计学意义,P=0.002;癌旁组织与正常组差异无统计学意义,P=0.609。大鼠肝癌组织Cyclin B1mRNA的表达量(0.002 8±0.001 9)高于正常肝组织(0.000 6±0.000 3),差异有统计学意义,P=0.003;其对应的癌旁组织Cyclin B1mRNA的表达量(0.002 9±0.002 5)高于正常肝组织,差异有统计学意义,P=0.001;肝癌组织与其对应的癌旁组织差异无统计学意义,P=0.809。树鼩肝癌组织Cyclin B1mRNA的表达量(0.014 8±0.010 9)高于正常肝组织(0.001 3±0.000 9),差异有统计学意义,P=0.007;对应癌旁组织Cyclin B1mRNA的表达量(0.008 3±0.006 4)高于正常肝组织,P=0.060;肝癌组织与对应的癌旁组织比较,差异无统计学意义,P=0.352。Cyclin B1mRNA在人肝癌组织中的表达与临床分期、门脉癌栓、术后复发、肿瘤直径及肿瘤分化明显有关,而与肝外转移、肿瘤个数及血清AFP水平无明显关系。Cyclin B1蛋白在人的肝癌组织中的平均表达量(0.570 7±0.221 9)明显高于其对应癌旁组织(0.418 5±0.254 3),差异有统计学意义,P=0.005;高于正常肝组织(0.375 4±0.127 0),差异有统计学意义,P=0.019。Cyclin B1在大鼠的肝癌组织中的平均表达量(0.661 9±0.398 1)明显高于正常肝组织(0.372 6±0.104 8),差异有统计学意义,P=0.039。Cyclin B1在树鼩的肝癌组织中的平均表达量(0.656 OBJECTIVE:To study the expression of Cyclin Bl(Ccnbl) in hepatocellular carcinoma (HCC) of differ ent species including human, rat and tree shrew (tupaia), and to verify the feasibility of cross-species oncogenomics ap-proach. METHODS: Real-time fluorescent quantitative polymerase chain reaction (Q-PCR) and Western Blot were applied to detect the expression levels of Cyelin B1 mRNA and protein levels in human(38 human HCC and Para-HCC tissues and 10 normal liver tissues),rat(15 HCC and Para-HCC tissues and 14 normal liver tissues),tree shrew(10 human HCC and para-HCC tissues and normal liver tissues),respectively. RESULTS: The expression of Cyelin B1 mRNA in HCC tissues of human(0. 011 4±0. 009 6) was significantly higher than that in normal liver tissues(0. 004 8+0. 002 9,P=0. 033) ; while in HCC adjacent tissues of human(0. 003 7± 0. 002 7) was significantly lower than that in HCC tissues (P = 0. 002). There was no significant difference between HCC-adjacent tissues and normal liver tissues (P=0. 609). The ex- pression of Cyclin B1 mRNA in HCC tissues of rat(0. 002 8±0. 001 9) was significantly higher than that in normal liver tissues(0. 000 6 ± 0. 000 3, P= 0. 003) ; while in HCC-adjaeent tissues of human(0. 002 9 ± 0. 002 5) was significantly higher than that in normal liver tissues (P= 0. 001). There was no significant difference between HCC-adjacent tissues and HCC tissues (P=0. 809). The expression of Cyclin B1 mRNA in HCC tissues of tree shew(0. 014 8±0. 010 9) wassignificantly higher than that in normal liver tissues(0. 001 3±0. 000 9 ,P=0. 007)while in HCC-adjaeent tissues of hu man(0. 008 3±0. 006 4) it was significantly higher than that in normal liver tissues (P=0. 060). There was no significant difference between HCC-adjacent tissues and HCC tissues (P= 0. 352).
