摘  要:目的探讨胡黄连苷Ⅱ在大鼠脑缺血-再灌注损伤中的抗氧化作用。方法应用线栓法建立MCAO-R模型,随机分为假手术组、阴性对照组、阳性对照组及药物治疗组。假手术组不做处理,阴性对照组经尾静脉注射生理盐水,阳性对照组注射丹参素钠10mg.kg-1,治疗组注射胡黄连苷Ⅱ10mg.kg-1。用Bederson法进行神经功能评分,氯化三苯基四氮唑(TTC)染色法测定脑梗死体积,TUNEL法检测细胞凋亡,免疫组化检测诱导型一氧化氮合酶(iNOS)蛋白表达变化,原位杂交检测诱导型iNOS mRNA表达变化。结果胡黄连苷II组神经行为功能、脑梗死体积、TUNEL阳性细胞数均低于阴性对照组(P<0.05),iNOS蛋白和mRNA表达低于阴性对照组(P<0.05)。结论胡黄连苷Ⅱ可能通过下调iNOS表达,对脑缺血-再灌注损伤有一定的保护作用。Objective To study the antioxidant effects of picroside ]I on the cerebral ischemia reperfusion injury in rats. Methods The MCAO/R models were established with a suture blocking in the MCA and were randomly divided into sham operative group, negative control group, positive control group and drug treatment group. The rats in the sham operative group were not treated, normal saline were injected into those in the negative control group, salvianic acid A sodium (10 mg/kg) were injected into those in the positive control group, and picroside 11 ( 10 mg/kg) were injected into those in the drug treatment group. The nervous behavioral function was evaluated with Bederson's test. The cerebral infarction volume was observed with tetrazolium chloride (rlTC) staining. TUNEL positive cells were counted by immunofluoresence assay. The expressions of inducible nitric oxide synthase (iNOS) was determined by immunohistochemical assay. The expressions of inducible nitric oxide synthase (iNOS) mRNA was determined by situ hybridization assay. Results In picroside II group rat, the nervous behavioral malfunction, the cerebral infarction volume and TUNEL positive cells were significantly lower than those in negative control group (P 〈 0.05). The expression and concentration of iNOS in picroside I1 group rat were significantly lower than those in the negative control group( P 〈 0.05). Conclusion Picroside 11 may reduce the expression of iNOS to inhibit the neuronal apoptosis, so that to play a neuroprotective role in cerebral artery occlusion and treperfusion injury in rats.
